  1. 1. Advanced molecular tools for diagnostic analyses of RNA and antibodies in situ and in solution

    Författare :Mengqi Wang; Ulf Landegren; Anders Ståhlberg; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Homogenous serological assay; PCR-based antibody detection; Proximity extension assay; SARS-CoV-2 antibody; Extracellular vesicles; Proximity barcoding assay; in situ superRCA; mutant transcript;

    Sammanfattning : Advanced molecular diagnostics uses in vitro biological assays to detect nucleic acids or proteins even in low concentrations across samples, allowing for the identification of biomarkers, monitoring the course of the disease over time, and selection of appropriate therapy. In this thesis, I focus on development and early applications of several molecular tools of expected value in research, and eventually also clinically. LÄS MER

    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; transcriptome reconstruction; transcriptome assembly; transcript isoform detection; Picea abies; acrocona; cone setting; differential expression; gene families; miRNA; quality assessment; transkriptomrekonstruktion; sammansättning av transkriptom; detektering av transkriptisoform; rödgran; kottegran; kottsättning; differentiellt uttryck; genfamiljer; kvalitetsutvärdering; miRNA; Biotechnology; Bioteknologi;

    Sammanfattning : Transcriptome reconstruction is an important component in the bioinformatical part of transcriptome studies. When a reference genome is missing, highly fragmented or incomplete, a de novo transcriptome assembly is the transcriptome reconstruction approach of choice, since in such situations, a simple alignment (or mapping) would not necessarily give all theinformation concerning splice junctions, isoforms or even the full extent of the gene. 3. 3. Methods for transcriptome reconstruction, with an application in Picea abies (L.) H. Karst

    Författare :Karl Johan Westrin; Olof Emanuelsson; Björn Nystedt; KTH; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; acrocona; cone setting; transcript isoform detection; miRNA; Picea abies; quality assessment; transcriptome assembly; Bioteknologi; Biotechnology;

    Sammanfattning : Transcriptome reconstruction is an important component in the bioinformatical part of transcriptome studies. It is particulary interesting when a reference genome is missing, highly fragmented or incomplete, since in such situations, a simple alignment (or mapping) would not necessarily tell the full story. 4. 4. Hypothalamic neuronal circuits involved in the regulation of food intake and body weight : histochemical studies in lean rats and obese mutant mice

    Författare :Matilda Bäckberg; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :AGRP; arcuate nucleus; BBB; CART; GABA; GABA receptors; immunohistochemistry; in situ hybridization; hypothalamus; LHA; leptin; MCH; NPY; orexin; POMC; tubby;

    Sammanfattning : Obesity is a major health problem because of its dramatic increase in prevalence and its association with type 2 diabetes, coronary heart diseases, hypertension and stroke. It is well known that signals generated in response to feeding behavior are integrated in the brain. LÄS MER

  5. 5. Neuropeptide circuitries regulating food and water intake

    Författare :Christian Broberger; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :agouti gene-related protein; anorexia mutation; arcuate nucleus; cholecystokinin; cocaine- and amphetamine-regulated transcript; coexistence; confocal microscopy; dorsal root ganglion; feeding; hypocretin; immunohistochemistry; in situ hybridization; melanin-concentrating hormone; nodose ganglion; nucleus tractus solitaxii; orexin; paraventricular nucleus; satiety; sensory neurones; pro-opiomelanocortin; vagus nerve;

    Sammanfattning : An adequate supply of nutrients through food intake is critical for survival, as evidenced by disorders such as obesity and anorexia, which in the long run may be life-threatening. Feeding behaviour is ultimately controlled by interacting neuronal populations in the brain. LÄS MER
