    Sammanfattning : Myocardial ischaemia initiates complex severe cellular changes and, after a critical time period, myocardial cell death. Despite that restitution of coronary blood flow is an absolute pre-requisite for tissue survival, myocardial reperfusion per se will also increase irreversible damage in the jeopardised myocardium.     Sammanfattning : The aims of this dissertation were to investigate the effects of extracellular purines on insulin secretion, and apoptosis in mouse pancreatic islets and β-cells; to examine if high glucose and free fatty acids induces β-cell apoptosis via autocrine effects of ADP acting on the P2Y13 receptor; and to investigate the modulation of extracellular purines in vascular smooth muscle cells (VSMCs) through ectonucleotidase or ATP release under the effect of high glucose. The expression of the ADP receptors P2Y1 and P2Y13 were shown in both mouse pancreatic islets and isolated β-cells using real-time PCR quantification. LÄS MER

    Sammanfattning : We have investigated the in vitro effects of long-chain free fatty acids (FFAs) on mouse pancreatic α- and β-cells by a combination of electrophysiological recordings, intracellular Ca2+ ([Ca2+]i) microfluorimetry and hormone release measurements. Acute addition of the saturated FFA palmitate to intact islets doubled insulin as well as glucagon secretion in the presence of 1 or 15 mM glucose and the FFA remained capable of enhancing release of both hormones in islets depolarised with high K+ in the presence of the KATP channel opener diazoxide, indicating that alterations of ATP sensitive potassium (KATP) channel activity is not part of the effect.     Sammanfattning : Neural responses to anoxia were studied in two fish species, the rainbow trout(Oncorhynchus mykiss) and the crucian carp (Carassius carassius). The rainbow troutbrain reacted to anoxia by a massive release of excitatory amino acids and K+ into theextracellular compartment, in a fashion very much like the brain of mammals duringanoxia.
