1. Roles of voltage-gated Ca2+ channel subunits in pancreatic β cells
Sammanfattning : Hallmarks of type 2 diabetes (T2D) include elevated blood glucose and free fatty acids (FFAs) as a result of impaired β cell insulin secretion and decreased β cell mass. The glucose-stimulated insulin secretion (GSIS) in β cells is triggered by depolarization-evoked Ca2+ entry through voltage-gated Ca2+ (CaV) channels. LÄS MER
2. Short- and long-term effects of long-chain free fatty acids on pancreatic α- and β-cell function
Sammanfattning : We have investigated the in vitro effects of long-chain free fatty acids (FFAs) on mouse pancreatic α- and β-cells by a combination of electrophysiological recordings, intracellular Ca2+ ([Ca2+]i) microfluorimetry and hormone release measurements. Acute addition of the saturated FFA palmitate to intact islets doubled insulin as well as glucagon secretion in the presence of 1 or 15 mM glucose and the FFA remained capable of enhancing release of both hormones in islets depolarised with high K+ in the presence of the KATP channel opener diazoxide, indicating that alterations of ATP sensitive potassium (KATP) channel activity is not part of the effect. 3. Temporal monitoring of intracellular Ca2+ signaling and origins of Ca2+ oscillations
Sammanfattning : This thesis examined parameters influencing stimulated cytoplasmic free Ca2+ concentration ([Ca2+]i) oscillations in hepatocytes and pancreatic beta-cells. Hepatic glucose output is regulated in part by hormones such as vasopressin that act through [Ca 2+]i oscillations. LÄS MER
4. Effects of imidazoline compounds on intracellular CA2+ and apoptosis
Sammanfattning : Cytoplasmic free Ca2+ concentration ([Ca2+]i) plays a pivotal role in the regulation of insulin release from the pancreatic beta-cell. [Ca2+]i is also important in the control of apoptosis. LÄS MER
5. Vacuolar Ca2+-ATPases in Plants. Regulation by calmodulin binding to a N-terminal autoinhibitory domain
Sammanfattning : The calcium ion (Ca2+) is essential for all plant and animal life. One important function of Ca2+ is as a second messenger in the responses of the cell to environmental and hormonal signals as well as in some intrinsic developmental processes. LÄS MER
