  1. 1. Pulsatile insulin release from single islets of Langerhans

    Författare :Johanna Westerlund; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Cell biology; islets of Langerhans; insulin; ELISA; cytoplasmic calcium; oscillations; type 2 diabetes; glucose; diazoxide; potassium; tolbutamide; DNP; antimycin A; iodoacetamide; microfluorometry; fura-2; KATP channels; Cellbiologi; Cell biology; Cellbiologi; medicinsk cellbiologi; Medical Cell Biology;

    Sammanfattning : Insulin release from single islets of Langerhans is pulsatile. The secretory activities of the islets in the pancreas are coordinated resulting in plasma insulin oscillations. Nutrients amplitude-regulate the insulin pulses without influencing their frequency. LÄS MER

  2. 2. Effects of over-stimulation by glucose on pancreatic beta-cell functioning : studies in vitro with diazoxide

    Författare :Anneli Björklund; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :diabetes mellitus; diazoxide; hyperglycemia; insulin secretion; islets of Langerhans; over-stimulation;

    Sammanfattning : Long term hyperglycemia impairs pancreatic ß-cell function. In vivo studies in rats have shown a decreased glucose-induced insulin response in the perfused pancreas after high glucose infusions. LÄS MER

  3. 3. Aspects on the modulation of potassium channels in insulin-producing beta-cells

    Författare :Zuheng Ma; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :islets of Langerhans; over-stimulation; diazoxide; exocytosis; proteasome; microarray; Kv1.1.;

    Sammanfattning : This thesis attempts to further clarify mechanisms behind the negative effects of overstimulation and the beneficial effect of β-cell rest. For this purpose diazoxide - opener of K+-ATP channels, which reversibly inhibits glucose-induced insulin secretion, was used as a probe. LÄS MER

  4. 4. The importance of immunological, genetic and clinical factors for beta cell function in childhood diabetes

    Författare :Eva Örtqvist; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Type I diabetes; HLA-DQ; GAD65Ab; 1A-2Ab; ICA; GAD65 epitopeC-peptide; clinical remission; beta cell rest; diazoxide.;

    Sammanfattning : Type I diabetes is caused by environmentally initiated, progressive autoimmune destruction of pancreatic beta cells in genetically susceptible individuals, which results in insulin deficiency and hyperglycemia. At diagnosis, about 10-20 % of beta cells remain, but after insulin therapy is started, a period with regained secretive capacity follows. LÄS MER

  5. 5. Endocrine aspects of obesity and weight reduction by bariatric surgery : with special emphasis on beta cell function

    Författare :Marie Guldstrand; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Obesity; weight reduction; b ariatric surgery; insulin; glucagon; type 2 diabetes; insulin sensitivity; HPA axis; autonomic nervous system ANS ; cognitive functions; proinsulin; GIP; GLP-1; IGFBP- 1; diazoxide;

    Sammanfattning : Severe obesity is the most predictive acquired risk factor for the development of type 2 diabetes. Bariatric surgery induces marked weight loss and reverses the diabetic state significantly. LÄS MER
