  1. 1. Harnessing the immunoregulatory capacity of single-stranded oligonucleotides to modulate innate immunity

    Författare :Aleksandra Dondalska; Anna-Lena Spetz; Jenny Hallgren Martinsson; Stockholms universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; single-stranded oligonucleotides; Toll-like receptors; influenza; respiratory syncytial virus; dendritic cell; mast cell; virus infection; inflammation; itch; molekylär biovetenskap; Molecular Bioscience;

    Sammanfattning : As the first line of defense, the innate immune system is exceptionally efficacious against invading pathogens but must be tightly regulated to avoid immune-related pathogenesis. Accordingly, the innate immune system recognizes highly conserved components associated with infections and damaged cells using a limited number of pattern-recognition receptors (PRRs), which are differentially expressed across cell types. 2. 2. The role of non-coding single-stranded oligonucleotides on Respiratory syncytial virus infection

    Författare :Sandra Axberg Pålsson; Anna-Lena Spetz; Annika Karlsson; Stockholms universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Respiratory syncytial virus RSV ; single-stranded oligonucleotide ssON ; small non-coding RNA sncRNA ; nucleolin; virus infection; antiviral; molekylär biovetenskap; Molecular Bioscience;

    Sammanfattning : Respiratory syncytial virus (RSV) is an enveloped RNA virus of the Pneumoviridae family. RSV is reported to infect host cells by receptor-mediated endocytosis, also called clathrin-dependent endocytosis. Many reports indicate that the virus utilizes the host receptor nucleolin for entry. 3. 3. Single-stranded DNA : methods and application in nanotechnology

    Författare :Giulio Bernardinelli; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :;

    Sammanfattning : Basic molecular and cell research, production of recombinant proteins, diagnostic detection of genetic mutations, construction of nanostructures and high-throughput DNA sequencing are only a few examples of the diverse set of applications that are amenable thanks to the availability of synthetic DNA polymers in biomedicine. Strategic investments and technical progress together with the introduction of automation in the synthesis of DNA oligomers enabled to transform, in just a few decades, a process mastered only by a niche of biochemists into an affordable and available custom-made product to every scientific field. 4. 4. Some Aspects of Physicochemical Properties of DNA and RNA

    Författare :Sandipta Acharya; Jyoti Chattopadhyaya; Piet Herdewijn; Uppsala universitet; []
    Nyckelord :Bioorganic chemistry; nucleic acids; hydrogen-bonding; stacking; single-stranded; NMR; molecular dynamics; Bioorganisk kemi;

    Sammanfattning : This thesis is based on nine research publications (I – IX) on structure and reactivity of RNA vis-à-vis DNA. The DNA and RNA are made of flexible pentose sugar units, polyelectrolytic phosphodiester backbone, and heterocyclic nucleobases. DNA stores our genetic code, whereas RNA is involved both in protein biosynthesis and catalysis. LÄS MER

  5. 5. Conformationally Constrained Oligonucleotides for RNA Targeting

    Författare :Qing Li; Jyoti Chattopadhyaya; Li-He Zhang; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; conformationally constrained nucleoside; antisense oligonucleotide; RNA affinity; nuclease stability; RNase H; RNA degradation; Chemistry with specialization in Bioorganic Chemistry; Kemi med inriktning mot bioorganisk kemi;

    Sammanfattning : A short oligonucleotide sequence as in a single-stranded antisense oligo nucleotides (AON) or in double-stranded small interfering RNAs (siRNA) can modulate the gene expression by targeting against the cellular mRNA, which can be potentially exploited for therapeutic purposes in the treatment of different diseases. In order to improve the efficacy of oligonucleotide-based drugs, the problem of target affinity, nuclease stability and delivery needs to be addressed. LÄS MER
