1. Lifestyle, biomarkers and the risk of developing inflammatory bowel disease
Sammanfattning : Introduction: Inflammatory bowel disease (IBD), including ulcerative colitis (UC) and Crohn’s disease (CD), is a chronic disease causing inflammation in the gut mucosa. The pathogenesis involves alteration in gut microbiota and in the intestinal barrier due to genetic factors, environmental exposure and dysregulation of the immune response. LÄS MER
2. Enzymatic mechanisms in biotin synthesis : vitamin B6 catalysis and phosphoryl transfer
Sammanfattning : With the large number of pathogenic bacteria that have developed antibiotic resistance it is increasingly important to find new targets for antibiotics. Our interest in the biotin biosynthesis pathway originates from this drug design perspective. All organisms require biotin, but it is only synthesised in bacteria, in plants and in some fungi. 3. Quantum Chemical Studies of Enzymatic Reaction Mechanisms : Investigations of Cytosine Deaminase and ω-Transaminase
Sammanfattning : In this thesis, density functional theory is used to study the reaction mechanisms of two dierent enzymes. Quantum chemical cluster models of the active sites were designed using available crystal structures. LÄS MER
4. Stability and inactivation mechanisms of two transaminases
Sammanfattning : In the past decades, more and more enzymes are employed as biocatalysts in industrial processes because of their advantages, such as high efficiency, substrate selectivity and stereoselectivity. Among them, amine transaminases (ATAs) are pyridoxal 5’-phosphate (PLP) dependent enzymes. LÄS MER
5. Structural Studies of Polyamine Biosynthesis Enzymes: Potential Targets for Drugs against Tropical Parasites
Sammanfattning : Two enzymes involved in the biosynthesis of polyamines, ornithine decarboxylase (ODC, from human and Leishmania donovani) and spermidine synthase (SPDS, from Plasmodium falciparum and Caenorhabditis elegans) were studied using biochemical methods, homology modelling and X-ray crystallography. ODC catalyses the first and committed step in polyamine biosynthesis, the decarboxylation of ornithine to putrescine. LÄS MER
