11. Cell-penetrating peptides and bioactive cargoes : Strategies and mechanisms
Sammanfattning : The cell membrane is an impermeable barrier for most macromolecules. Recently discovered cell-penetrating peptides (CPPs) have gained lot of attention because they can cross the membrane, and even more, carry cargoes with them. How CPPs enter cells is still not clear, while the delivery of different cargoes has been convincingly shown. LÄS MER
12. Isolation, structure and processing of bioactive peptides
Sammanfattning : Amino acid sequences deduced from DNA sequences do not prove in which forms proteins, peptides and their processed products are present in the tissues. The products of a single gene may give rise to variants of peptides because of post-transcriptional or post-translational processing. LÄS MER
13. Complexes of cell-penetrating peptides with oligonucleotides : Structure, binding and translocation in lipid membranes
Sammanfattning : The fundamental element of life known to man is the gene. The information contained in genes regulates all cellular functions, in health and disease. The ability to selectively alter genes or their transcript intermediates with designed molecular tools, as synthetic oligonucleotides, represents a paradigm shift in human medicine. LÄS MER
14. Rational design of novel cell-penetrating peptides
Sammanfattning : Although cell-penetrating peptides (CPPs) have been an active field of research for over 20 years there are still properties such as efficiency and stability that must be improved for CPPs to reach their full potential as delivery vectors. In this thesis two peptides were designed for non-covalent oligonucleotide delivery, in an attempt to overcome two major issues hampering wider use of CPPs, namely low stability and endosomal entrapment. 15. Vectorization of oligonucleotides with cell-penetrating peptides : Characterization of uptake mechanisms and cytotoxicity
Sammanfattning : The hydrophobic plasma membrane constitutes an indispensable barrier for cells in living animals. Albeit being pivotal for the maintenance of cells, the inability to cross the plasma membrane is still one of the major obstacles to overcome in order to progress current drug development. LÄS MER
