6. Chemoenzymatic Resolution in Dynamic Systems : Screening, Classification and Asymmetric Synthesis
Sammanfattning : This thesis is divided into four parts, all centered around Constitutional Dynamic Chemistry (CDC) and Dynamic Kinetic Resolution (DKR) using biocatalysts for selective transformations, and their applications in screening of bioactive compounds, organic synthesis, and enzyme classification. In part one, an introduction to CDC and DKR is presented, illustrating the basic concepts, practical considerations and potential applications of such dynamic systems, thus providing the background information for the studies in the following chapters. 7. Directed Evolution of Glutathione Transferases with Altered Substrate Selectivity Profiles : A Laboratory Evolution Study Shedding Light on the Multidimensional Nature of Epistasis
Sammanfattning : Directed evolution is generally regarded as a useful approach in protein engineering. By subjecting members of a mutant library to the power of Darwinian evolution, desired protein properties are obtained. Numerous reports have appeared in the literature showing the success of tailoring proteins for various applications by this method. LÄS MER
8. Engineered Alcohol Dehydrogenases for Stereoselective Chemical Transformations
Sammanfattning : Enzymes are biomolecules built from amino acids and catalyze the chemical transformations in a cell. Enzymes are by nature stereoselective, biodegradable, environmentally friendly, and can perform catalysis in aqueous solutions and at ambient temperatures. LÄS MER
9. Exploring Conjugate Addition Activity in Pseudozyma antarctica Lipase B
Sammanfattning : Multifunctional enzymes have alternative functions or activities, known as “moonlighting” or “promiscuous”, which are often hidden behind a native enzyme activity and therefore only visible under special environmental conditions. In this thesis, the active-site of Pseudozyma (formerly Candida) antarctica lipase B was explored for a promiscuous conjugate addition activity. LÄS MER
10. Rational engineering of esterases for improved amidase specificity in amide synthesis and hydrolysis
Sammanfattning : Biocatalysis is an ever evolving field that uses enzymes or microorganisms for chemical synthesis. By utilizing enzymes that generally have evolved for specific reactions under mild conditions and temperatures, biocatalysis can be a more environmentally friendly option compared to traditional chemistry.
