21. Neurorestorative strategies involving neurogenesis, neuronal precursors and stem cells in animal models of Parkinson's disease
Sammanfattning : The general aims of the thesis is to provide evidence for neurogenesis in the adult substantia nigra pars compacta (SNpc), where the dopamine-producing neurons lost in Parkinson's disease (PD) reside; to optimize methods used for detecting newborn nerve cells in adult brain regions with a low rate of neurogenesis; to explore the mechanism of nerve cell death in animal models of parkinsonism and early degenerative and restorative changes in nigral cell populations; and to develop a therapeutic approach that could translate into a future restorative disease-modifying strategy in PD. 22. Brain-derived neurotrophic factor in cerebral ischemia: a quantitative study on surviving and newly formed neurons
Sammanfattning : This study explores the role of brain-derived neurotrophic factor (BDNF) for neuronal vulnerability and neurogenesis following cerebral ischemia in the rat. Cerebral ischemia results in selective neuronal cell loss in specific brain regions. We here demonstrate the rank order of vulnerability in two regions affected by global forebrain ischemia, i. 23. Adult neurogenesis : from stem cell to functional neuron
Sammanfattning : The adult mammalian central nervous system harbors a population of neural stem cells with the ability to generate neurons, astrocytes and oligodendrocytes. These pluripotent cells can be enriched in vitro, and by directing their differentiation it would be possible to generate populations of specific neural cell types to use for transplantation. LÄS MER
24. A long and winding road: Differentiating human embryonic stem cells into dopaminergic neurons
Sammanfattning : Parkinson’s disease is a progressive neurodegenerative disorder. Since the motor manifestations of the disease is associated with the loss of dopaminergic (DAergic) neurons in the substantia nigra pars compacta, cell replacement therapy is a promising approach for treatment. 25. Mechanisms and time course of cell death in embryonic nigral tissue and intrastriatal transplants
Sammanfattning : Parkinson’s disease (PD) is the second most common neurodegenerative disorder and severely affects about one in a hundred persons over the age of fifty years. In PD, there is a reduced striatal concentration of the neurotransmittor dopamine, mainly caused by a degeneration of the dopaminergic neurons in the substantia nigra. LÄS MER
