1. Bioprocess Monitoring and Control in Pseudomonas cepacia and recombinant Escherichia coli cultivations
Sammanfattning : The control of cultivation conditions and its influence on growth and production have been investigated. The use of fed-batch processes have been demonstrated for several cultivation systems and so has the use of different control signals in these systems. LÄS MER
2. Growth rate control of periplasmic product retention in Escherichia coli
Sammanfattning : The recombinant product is secreted to the periplasm in many processes where E. coli is used as host. One drawback with secretion is the undesired leakage of the periplasmic products to the medium. The aim of this work was to find strategies to influence the periplasmic retention of recombinant products. LÄS MER
3. Impact of glucose feed rate on productivity and recombinant protein quality in Escherichia coli
Sammanfattning : The goal of this work was to contribute to the fed-batch process optimisation task by deriving parameters that have considerable impact on productivity as well as product quality The chosen parameters were I) the design of the glucose feed profile, II) the choice of induction strategy, with respect to the method of addition, and III) the time of the induction, with respect to the specific glucose consumption rate. The present fed-batch experiments using the lacUV5-promoter, for production of b-galactosidase, have shown that a high glucose feed rate gives a specific production rate, qp, that is twice as high, after induction, compared to a feed rate that is 2. 4. Impact of glucose uptake rate on recombinant protein production in Escherichia coli
Sammanfattning : Escherichia coli (E. coli) is an attractive host for production of recombinant proteins, since it generally provides a rapid and economical means to achieve high product quantities. LÄS MER
5. Bench-Scale Production of Heterologous Proteins from Extremophiles- Escherichia coli and Pichia pastoris based expression systems
Sammanfattning : Over the past few years considerable research attention has been assigned to extremophiles as sources of extremozymes due to their applicability in industrial processes, and the development of eco-friendly technologies. The establishment of efficient production strategies for heterologous proteins is an empirical process requiring broad background knowledge on available expression systems together with their major advantages and shortcomings.
