  1. 1. Engineering yeast for improved recombinant protein production

    Författare :Yanyan Wang; Chalmers tekniska högskola; []
    Nyckelord :TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; NATURVETENSKAP; NATURAL SCIENCES; point mutation; genome-scale modeling; recombinant protein; omics analysis; CRISPR Cas9; protein secretion; Saccharomyces cerevisiae;

    Sammanfattning : Recombinant proteins are broadly used from basic research to therapeutic development and include industrial enzymes and pharmaceutical proteins. The increasing demand for improved production and enhanced quality of recombinant proteins requires robust biotech-based strategies to overcome the limitations of protein extraction from natural sources. 2. 2. Protein production, characterization and structure determination in structural genomics

    Författare :Esmeralda A. Woestenenk; Torleif Härd; Göran Karlsson; KTH; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Biology; protein production; protein solubility; fusion tags; nuclear magnetic resonance; structure determination; Biologi; Biology; Biologi;

    Sammanfattning : This thesis covers the process from expression of a heterologous gene in Escherichia coli to structure determination of a protein by nuclear magnetic resonance (NMR) spectroscopy. The first part concerns structural genomics-related parallel screening studies on the effect of fusion tags (in particular the His tag) on protein solubility and the use of fusion tags in fast, parallel purification protocols intended for initial biophysical characterization of human proteins produced in E. 3. 3. Systems Analysis of the Protein Secretory Pathway in Yeast and Human Cells

    Författare :Amir Feizi; Chalmers tekniska högskola; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; protein production; flux balance analysis; biopharmaceutical; secretory pathway: secretome; recombinant protein; genome-scale modeling; systems biology;

    Sammanfattning : The cell was discovered by Robert Hooke in 1665, later in nineteen century the cell theory was developed as all organisms are composed of one or more cells. Since then, many tools and experimental methods have been developed to dissect and characterize the cell components which resulted in established solid scientific fields such as biochemistry, molecular cell biology and genetics. 4. 4. Bench-Scale Production of Heterologous Proteins from Extremophiles- Escherichia coli and Pichia pastoris based expression systems

    Författare :Santosh Ramchuran; Bioteknik; []
    Nyckelord :TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; Glycoside hydrolase; Heterologous protein; High cell density cultivation Expression systems; Bioteknik; Biotechnology; Fed-batch;

    Sammanfattning : Over the past few years considerable research attention has been assigned to extremophiles as sources of extremozymes due to their applicability in industrial processes, and the development of eco-friendly technologies. The establishment of efficient production strategies for heterologous proteins is an empirical process requiring broad background knowledge on available expression systems together with their major advantages and shortcomings. 5. 5. Metabolic Engineering of Recombinant Protein Production by Saccharomyces cerevisiae

    Författare :Zihe Liu; Chalmers tekniska högskola; []
    Nyckelord :TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; recombinant protein production; genome sequencing; DNA microarray; insulin precursor; growth rate; UV mutation; yeast; anaerobic electron acceptor; secretory pathway; unfolded protein response; metabolic engineering; systems biology; α-amylase;

    Sammanfattning : The yeast Saccharomyces cerevisiae is a widely used cell factory for the production of fuels, chemicals, and it also provides a platform for the production of many heterologous proteins of medical or industrial interest. In this thesis, random and rational approaches, such as vector design, host engineering, fermentation analysis, UV Mutation, coupled with high-throughput systems biology techniques (including whole genomic sequencing, microarray analysis and flux analysis) and integrated analysis (Reporter feature technique), were employed to engineer cellular properties more effectively and purposefully to construct cell factories for protein production.
