  1. 1. Expanding the alphabet of fluorescent nucleic acid base analogues: Characterization of new members

    Författare :Anke Dierckx; Chalmers tekniska högskola; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; fluorescent adenine analogue; fluorescent DNA base analogue; duplex stability; Fluorescence quantum yield; circular dichroism; B-DNA;

    Sammanfattning : Fluorescence has proven to be a powerful tool for studying structure, dynamics and interactions of biomolecules such as proteins, RNA and DNA. However, since the natural DNA/RNA nucleobases are virtually non-fluorescent, there is a need for developing fluorescent nucleic acid markers. LÄS MER

  2. 2. Fluorescent Nucleobase Analogues and their use for Investigating DNA Interactions

    Författare :Anke Dierckx; Chalmers tekniska högskola; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; FRET; B-DNA; circular dichroism; fluorescent nucleobase analogue; adenine analogue; duplex stability; Fluorescence quantum yield;

    Sammanfattning : Ever since unravelling the structure of DNA, an expanding research field has emerged with ongoing efforts dedicated to increase our understanding of the molecule of life. Since the natural nucleobases are virtually non-emissive, it has been a challenge for decades to ‘light up’ DNA/RNA in order to investigate their properties utilizing fluorescence techniques. 3. 3. Development of benzoquinoquinoxaline derivatives as triplex-specific probes : Recognition of DNA structures at repeats sequences

    Författare :Helen Bergquist; Rula Zain; Keith Fox; Stockholms universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Triplex; H-DNA; BQQ; BQQ-OP; BQQ-Bodipy; triplet repeat; DNA; non-B-DNA; pkd1; frataxin; Friedreich s ataxia; Molecular biology; Molekylärbiologi; Molecular Biology; molekylärbiologi;

    Sammanfattning : Repeat sequences are associated with several human diseases, such as Friedreich’s ataxia, polycystic kidney disease and cancer. These sequences can form non-B-DNA structures, including triplex (H-DNA) DNA, and are associated with genomic instability and altered gene expression. LÄS MER

  4. 4. Structural Adaptation of DNA to Small Ligands and Recombination Proteins

    Författare :Hye-Kyung Kim; Chalmers tekniska högskola; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; triplex DNA; groove binders; intercalators; SANS; DNA; optical spectroscopy; recombination proteins;

    Sammanfattning : In order to contribute to the understanding of mechanisms of recognition in nucleic acid systems, the interaction of DNA with a variety of ligands, representing different cases of adaptation of the double-helix structure, was studied using flow linear dichroism (LD), circular dichroism (CD), fluorescence and absorption spectroscopic techniques. A chiral hydrophobic metal complex, [Ru(dpphen)3]2+ was examined, since this had been reported to be the most efficient molecule to discriminate DNA handedness. LÄS MER

  5. 5. DNA strand exchange and hydrophobic interactions between biomolecules

    Författare :Bobo Feng; Chalmers tekniska högskola; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; ATP synthase; DNA; membrane proteins; linear dichroism; Hydrophobic interactions; self-assembly; strand exchange; nanotechnology; liposomes;

    Sammanfattning : The role of hydrophobic interactions in DNA strand exchange has been studied using fluorescence-labeled DNA oligomers in a FRET assay. Strand exchange was found to be accelerated in the presence of polyethylene glycol, which provides a crowded and hydrophobic environment possibly mimicking that of the catalytically active recombinase-DNA complexes.
