1. Transglycosylation catalyzed by β-mannanases : Molecular aspects relevant for enzymatic synthesis
Sammanfattning : β-Mannanases are a major type of enzyme involved in the hydrolysis and modification of β-mannans, a hemicellulose present in high amounts in e.g. softwoods such as spruce. The β-mannanases studied in this thesis belong to glycoside hydrolase (GH) families GH5 and GH26. LÄS MER
2. β-Mannoside hydrolases from clan A – Structural enzymology, subsite engineering and transglycosylation
Sammanfattning : β-Mannoside hydrolases are enzymes that are involved in conversion of the major soft-wood hemicellulose O-acetyl-galactoglucomannan, an important renewable biomass resource. They belong to clan A of glycoside hydrolases and share a common (β/α)8-TIM barrel fold. LÄS MER
3. Enzymatic conversion of β-mannans: Analysing, evaluating and modifying transglycosylation properties of glycoside hydrolases
Sammanfattning : Retaining glycoside hydrolases are enzymes that catalyse the breakdown down of glycans through hydrolysis. Due to the double-replacement mechanism of the retaining glycoside hydrolases (GHs), which form an intermediate with part of the glycan covalently attached to the enzyme, some GHs are able to catalyse synthesis reactions called transglycosylation. 4. Structure and Function of Beta-Mannanases from Glycoside Hydrolase Clan A. A Study of Hemicellulases from Microbes and a Mollusc
Sammanfattning : Hemicellulose is the second most abundant group of renewable polysaccharides. Mannans can be found as the major hemicellulose in softwood and as storage polysaccharides in a range of plants. In this work, structural and functional characterisations of family 5 and 26 endo-1,4-b-mannanases (b-mannanases) have been performed. LÄS MER
5. Heterologous expression and characterization of xyloglucan endotransglycosylases from Populus tremula x tremuloides
Sammanfattning : Xyloglucan endotransglycosylases (XETs) are enzymes involvedin the reorganization of the primary cell wall. This thesisdescribes several aspects of XET16A from hybrid aspen (Populustremula x tremuloides). The gene was isolated from the hybridaspen cDNA library and the protein was expressed heterologouslyin the methylotrophic yeast Pichia pastoris. LÄS MER
