1. Label-free processing of stem cell preparations by acoustophoresis
Sammanfattning : The first bone marrow transplant in a human was performed in 1959, providing evidence that cells can be used for transplantation and treatment, revealing that healing capacities lie within the human body that ought to be understood. Since then hematopoietic stem cell transplantation has developed as a standard treatment for many cancers besides other malignancies. LÄS MER
2. Pathways that govern hematopoietic stem cell fate
Sammanfattning : Hematopoietic stem cells (HSCs) compose a rare population of undifferentiated cells, residing in the bone marrow of adult individuals, ensuring life-long maintenance and replenishment of the blood system. This fantastic achievement is possible owing to two special characteristics of the HSCs: their ability to make copies of themselves (self-renew), and their capacity to differentiate to all lineages of the blood system. 3. Development of Gene Therapy for Hematopoietic Stem Cells using Lentiviral Vectors
Sammanfattning : Hematopoietic stem cells are an ideal target for genetic manipulation for the purpose of curing hematological disorders as they have the ability to develop into all blood lineages and to self-renew. In this study we demonstrate that lentiviral vectors, based on HIV-1, can efficiently transfer genes into human hematopoietic progenitor and stem cells as assessed following stem cell transplantation in immune compromised mice. 4. Cell and gene therapy for blood disorders. Modeling of novel treatment strategies for Gaucher type 1 disease and Diamond-Blackfan anemia in mice
Sammanfattning : Gaucher disease type 1 (GD type 1) and Diamond-Blackfan anemia (DBA) are rare genetic disorders affecting the hematopoietic system and routinely treated by administration of drugs with variable clinical efficacy. Though effective in alleviating disease symptoms in large cohorts of patients, these treatment options have the major drawback of being non-curative. LÄS MER
5. Development of Novel Therapies, Models, and Biomarkers for Osteoclast-Related Diseases
Sammanfattning : Osteoclasts are derived from hematopoietic stem cells (HSCs) and form upon stimulation of osteoclast precursors by macrophage colony-stimulating factor (M-CSF) and receptor activator of nuclear factor κΒ ligand (RANKL). Osteoclasts resorb bone by secreting hydrochloric acid, matrix metalloproteinases (MMPs) and cathepsin K. LÄS MER
