11. Neogenesis and alloxan toxicity in pancreatic islets, with special reference to the transplanted human β-cell
Sammanfattning : The aim of this work was to study whether adult human pancreatic β-cells have the capacity to proliferate in culture and after transplantation to the renal subcapsular space of mice. An immunosuppressive regimen for human islet discordant xenotransplantation to mice was designed. LÄS MER
12. Mitochondrial medicine. New strategies to evaluate drug toxicity and develop pharmacological protection of the cell’s powerhouse
Sammanfattning : Mitochondria produce the majority of the cell’s energy. Any dysfunction in, or interference with mitochondrial function can have severe consequences. And yet, it was only within the last decades that screening for potential mitochondrial toxicity was included as routine toxicity assay during early drug development. LÄS MER
13. Cell models for evaluation of adult and developmental neurotoxicity : Focus on acrylamide
Sammanfattning : This thesis is aimed at summarizing some of the alternative in vitro methods and models that have been used to study both adult and developmental neurotoxicity (DNT), and also to pinpoint some of the important aspects of using alternative in vitro methods. The aim of the papers included in this thesis was to challenge the hypothesis that neurotoxicity and DNT of chemicals can be studied using robust endpoints for proliferation and neural differentiation, such as neurite outgrowth, mRNA expression and protein expression, in two different cell lines. 14. Acute lung failure induced by tricyclic antidepressants : an experimental evaluation
Sammanfattning : The tricyclic antidepressant drugs (TCA) have, over the years, been reported to be able to cause adult respiratory distress syndrome (ARDS)/acute lung injury in connection with overdose, and are currently among the most common cause of self-poisoning. Approximately 10% of all TCA overdose patients develop ARDS, and the treatment of these patients, irrespective of the event causing ARDS, is mainly supportive. LÄS MER
15. Optimization of in vivo cell tracking methods with long-lived radionuclides for PET imaging
Sammanfattning : Stem cell transplantation and cell therapy are the standard treatments for many diseases; however, many suffer from complications, and the therapeutic efficiency can be challenging to predict. The off-target accumulation of the transplanted cells can cause some of these shortcomings. LÄS MER
