  1. 6. Bombesin Antagonists for Targeting Gastrin-Releasing Peptide Receptor-Positive Tumors : Design, Synthesis, Preclinical Evaluation and Optimization of Imaging Agents

    Författare :Zohreh Varasteh; Anna Orlova; Roger Schibli; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Bombesin; Gastrin-releasing peptide receptor GRPR ; Antagonist; Radionuclide molecular imaging;

    Sammanfattning : This thesis is focused on the development, preclinical evaluation, and optimization of radiotracers for the detection of gastrin-releasing peptide receptor (GRPR)-expressing tumors. The work is divided into three distinct parts: (1) the development of bombesin (BN) antagonist (RM26)-based imaging radiotracers for the detection of GRPR-expressing tumors using different positron emission tomography (PET) and single photon emission computed tomography (SPECT) radionuclides (68Ga, 18F and 111In), (2) the establishment of a method to monitor the ligand-G protein-coupled receptor (GPCR) interaction in real time without requiring purification and stabilization of the receptors, and (3) the evaluation of radiopeptide structure-related factors (length of mini-PEG linker and composition of chelator for metal labeling) affecting the in vitro and in vivo characteristics of RM26-based tracers. 2. 7. The neuropeptides GRP and PACAP and the function of the endocrine pancreas - A study on receptor deficient mice

    Författare :Kristin Persson; Lund Medicin; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Endocrinology; secreting systems; diabetology; Endokrinologi; sekretion; hypoglycemia; glucagon; insulin; islet of Langerhans; diabetes; PACAP; neuropeptides; GRP; diabetologi;

    Sammanfattning : The parasympathetic nerves are important for the regulation of insulin secretion, in particular after meal intake. Apart from acetylcholine, several neuropeptides can be found in the parasympathetic nerves innervating the islets of Langerhans, including gastrin-releasing peptide (GRP) and pituitary adenylate cyclase activating polypeptide (PACAP). 3. 8. Signaling Mechanisms in the Neuronal Networks of Pain and Itch

    Författare :Katarzyna Rogoz; Klas Kullander; Malin Lagerström; Stephen McMahon; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; mouse genetics; neuronal network; glutamate; sensory neuron; dorsal root ganglion; pain; itch; Neuroscience; Neurovetenskap; Molekylär genetik; Molecular Genetics;

    Sammanfattning : Glutamate is the essential neurotransmitters in pain pathways. The discovery of the vesicular glutamate transporters (VGLUT1-3) has been a fundamental step on the way to describe glutamate-dependent pain pathways. We used the Cre-lox system to construct conditional knockouts with deficient Vglut2 transmission in specific neuronal populations. LÄS MER
