  1. 36. Engineering enzymes towards biotherapeutic applications using ancestral sequence reconstruction

    Författare :Natalie Hendrikse; Per-Olof Syrén; Johan Rockberg; Erik Nordling; Reinhard Sterner; KTH; []
    Nyckelord :TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; enzyme engineering; enzyme evolution; ancestral sequence reconstruction; therapeutic enzymes; terpene cyclases; ammonia-lyases; sulfatases; lysosomal enzymes; Kemi; Chemistry;

    Sammanfattning : Enzymes are versatile biocatalysts that fulfill essential functions in all forms of life and, therefore, play an important role in health and disease. One specific application of enzymes in life science is their use as biopharmaceuticals, which typically benefits from high catalytic activity and stability. LÄS MER

  2. 37. Oxidation of 1,2-Diols Using Alcohol Dehydrogenases : From Kinetic Characterization to Directed Evolution

    Författare :Cecilia Blikstad; Mikael Widersten; Stefan Lutz; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; enzyme kinetics; alcohol dehydrogenase; directed evolution; enzyme engineering; diol; α-hydroxy aldehyde; Biokemi; Biochemistry;

    Sammanfattning : The use of enzymes as catalysts for chemical transformations has emerged as a “greener” alternative to traditional organic synthesis. An issue to solve though, is that enzymes are designed by nature to catalyze reactions in a living cell and therefore, in many cases, do not meet the requirements of a suitable biocatalyst. LÄS MER

  3. 38. Probing the Alpha Class Glutathione Transferase Structure. Combinatorial Subunit Studies and Rational Redesign

    Författare :Ann Gustafsson; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Biochemistry; Glutathione transferases; rational redesign; enzyme; catalysis; Biokemi; Biochemistry; Biokemi; Molecular Biotechnology; molekylär bioteknik avd f jonfysik ;

    Sammanfattning : The present work concerns two Alpha class enzymes, GST A1-1 and GST A4-4, of the glutathione transferase (GST) family, which are detoxication enzymes catalyzing the conjugation of the tripeptide glutathione with various electrophiles. The GST A1-1 structure was explored for protein engineering purposes and tailored for increased activity toward alkenals. LÄS MER

  4. 39. Ethanol from lignocellulose : Management of by-products of hydrolysis

    Författare :Björn Alriksson; J Leif Jönsson; Vincent Eijsink; Karlstads universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Ethanol; Lignocellulose; Detoxification; Genetic engineering; Enzyme production; Biochemistry; Biokemi; Kemi; Chemistry;

    Sammanfattning : Fuel ethanol can be produced from lignocellulosic materials, such as residues from agriculture and forestry. The polysaccharides of lignocellulose are converted to sugars by hydrolysis and the sugars can then be fermented to ethanol using microorganisms. However, during hydrolysis a wide range of by-products are also generated. LÄS MER

  5. 40. Standardization of Islet Isolation and Transplantation Variables

    Författare :Andrew S Friberg; Olle Korsgren; Daniel Brandhorst; Paul R. V. Johnson; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Islet isolation; standardization; enzyme; gradient separation; digital imaging analysis; DIA; transplantation outcome; islet transplantation; prediction; Medical technology; Medicinsk teknik; Medical Cell Biology; Medicinsk cellbiologi; Computerized Image Processing; Datoriserad bildbehandling;

    Sammanfattning : Currently, the transplantation of islets of Langerhans is a viable means to maintain control of blood sugar levels and reduce the risk of hypoglycemia in defined populations with brittle type I diabetes mellitus or those requiring pancreatectomy. However, the process of islet isolation is highly variable and not all isolations result in islet numbers or quality suitable for transplantation. LÄS MER
