1. Adoptive T Cell Therapy of Viral Infection and Cancer : Ex vivo Expansion of Cytomegalovirus- and Prostate Antigen-specific T Cells
Sammanfattning : The main focus of my thesis has been to develop protocols for generating antigen-specific cytotoxic T lymphocytes (CTLs) and T helper cells (TH) for adoptive transfer to treat cytomegalovirus (CMV) disease and prostate cancer. CMV viremia is a severe complication in immunocompromised stem cell transplanted patients. LÄS MER
2. Glucocorticoid in T cell differentiation
Sammanfattning : Thymocytes differentiate into mature T cells with an immunological restriction for self major histocompatibility complex (MHC) antigens. Cells that do not recognize self MHC expressed on thymic epithelial cells (TEC) die by a rapid induction of apoptosis (default selection). LÄS MER
3. Smoking and T cell co-stimulation in rheumatoid arthritis
Sammanfattning : In this thesis I investigated if smoking limits the co-stimulatory system of CD8+ T cells in rheumatoid arthritis (RA). I took special interest in the co-inhibitory receptor PD-1 and its ligand PD-L1. LÄS MER
4. Regulatory mechanisms controlling T cell activity
Sammanfattning : This thesis is based on work studying the immunoregulatory mechanisms that govern the balance between activation and inactivation of T lymphocytes. Most of the work has been carried out in an experimental system using a T cell receptor (TCR-Vb3) transgenic mouse strain to obtain a homogenous T-cell population and utilizing the properties of bacterial superantigens to induce T cell responses and unresponsiveness in vivo. 5. Aspects of T cell development contributing to autoimmunity
Sammanfattning : Autoimmunity, such as type 1 diabetes (T1D), arises as a result of tolerance breakdown against host tissues, i.e. the β-cells. Central tolerance is established during thymic T cell development and defects in key events during this process could contribute to the pathogenesis of autoimmune diseases. LÄS MER
