  1. 1. Salvage and de novo synthesis of nucleotides in Trypanosoma brucei and mammalian cells

    Författare :Artur Fijolek; Lars Thelander; Arne Holmgren; Umeå universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; nucleotides; ribonucleotide reductase; Ara-A; p53R2; Trypanosoma brucei; African sleeping sickness; trypanosomiasis; CTP synthetase; adenosine kinase; acivicin; adenine arabinoside; Biochemistry; Biokemi;

    Sammanfattning : All living cells are dependent on nucleic acids for their survival. The genetic information stored in DNA is translated into functional proteins via a messenger molecule, the ribonucleic acid (RNA). Since DNA and RNA can be considered as polymers of nucleotides (NTPs), balanced pools of NTPs are crucial to nucleic acid synthesis and repair. LÄS MER

  2. 2. The Structure-Function Relationship and Evolution of Deoxyribonucleoside Kinases

    Författare :Zeeshan Mutahir; Molekylär cellbiologi; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES;

    Sammanfattning : The synthesis of deoxyribonucleoside triphosphates (dNTPs), building blocks of DNA, can be achieved by two different pathways in the cell. One is called de novo synthesis and the other is known as salvage pathway. The de novo pathway involves synthesis of dNTPs by utilizing basic molecules like sugars, amino acids, CO2, NH3, etc. LÄS MER

  3. 3. Deoxyribonucleoside kinases in bacteria, plants and humans

    Författare :Anders Ranegaard Clausen; Biologiska institutionen; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; anti-viral drugs; gene therapy; cancer; evolution; nucleoside analog; deoxyribonucleoside kinase; thymidine kinase;

    Sammanfattning : Deoxyribonucleoside triphosphates (dNTPs) are the building blocks of DNA and can be synthesized either de novo or via salvage pathways. The first reaction in the salvage pathway of dNTPs is the conversion of deoxyribonucleosides (dN) into the deoxyribonucleoside monophosphate (dNMP). LÄS MER

  4. 4. Studies of enzymes in mitochondrial DNA precursor synthesis : regulatory mechanisms for human thymidine kinase 2 and deoxyguanosine kinase

    Författare :Ren Sun; Sveriges lantbruksuniversitet; Sveriges lantbruksuniversitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES;

    Sammanfattning : As important enzymes in mitochondrial nucleotide salvage pathway, thymidine kinase 2 (TK2) and deoxyguanosine kinase (dGK) are expressed constitutively in almost all cells. These two enzymes catalyze the initial rate-limiting phosphorylation of pyrimidine and purine deoxynucleosides, respectively, providing DNA precursors for mitochondrial DNA (mtDNA) replication. 5. 5. Structural and functional studies of enzymes in nucleotide metabolism : a detailed investigation of two enzymes and interaction profiling of FDA-approved nucleoside analog drugs with 23 enzymes

    Författare :Louise Egeblad; Sveriges lantbruksuniversitet; Sveriges lantbruksuniversitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES;

    Sammanfattning : Enzymes in nucleotide metabolism serve as the producers of the building blocks for DNA and RNA. From a medical perspective, nucleotide metabolism, and in particular salvage pathway enzymes, have attracted special interest, as nucleoside prodrugs given in the treatment of cancer and HIV are converted into their active metabolite forms by these enzymes.
