6. Humoral immunity during primate lentivirus infection : evaluation of B cell apoptosis and engineering of virus specific antibodies
Sammanfattning : The focus of this project was to evaluate the contribution of the humoral immunity in controlling infection with primate lentiviruses - an area which has implications both for vaccine design as well as for understanding of the pathogenesis of the disease. The first goal was to generate efficient neutralizing reagents which could be used for passive immunotherapy trials, preferably in the macaque system, and to investigate if peptide immunogens could elicit such antibodies. 7. Dendritic cell and IgA responses in SIV and HIV-1 pathogenesis
Sammanfattning : Simian immunodeficiency virus (SIV) as well as human immunodeficiency virus (HIV-1) can infect different subsets of dendritic cells (DC). Such cells may therefore not only be involved in the generation of recruitment of naïve T and B lymphocytes in induction of SIV/HIV-1 specific adaptive responses but also transmit virus from port of entrance to the secondary lymphoid compartment generating systemic viral spread. Development of inexpensive inhibitors is necessary to allow treatment of HIV infections in the Third World.In the present thesis, the design and synthesis of several inhibitors of the HIV-1 virus are described. 9. Regulation of HIV-1 provirus transcription
Sammanfattning : Transcription of the HIV-genome is regulated by the interaction of viral long terminal repeat (LTR) cis-acting sequences with a complex of viral regulatory proteins and cellular transcription factors. The complex interplay between the cellular-, and/or viral proteins and LTR of different subtypes may play a role in the control of HIV-1 latency versus activation. LÄS MER
10. Development of lentiviral vectors for CNS gene transfer
Sammanfattning : Gene therapy in the brain is a promising treatment strategy that in the future may be used for several brain disorders, including Parkinson's disease, Alzheimer's disease and lysosomal storage diseases. By introducing a new gene, rather than providing a classical pharmacological drug, gene therapy offers the opportunity to treat neurological disorders by a single intervention. LÄS MER
