  1. 1. Epigenetic Dysregulations in the Brain of Human Alcoholics : Analysis of Opioid Genes

    Författare :Igor Bazov; Georgy Bakalkin; Catharina Lavebratt; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; neuropeptides; dynorphin; human brain; alcohol dependence; epigenetics; gene transcription;

    Sammanfattning : Neuropeptides are special in their expression profiles restricted to neuronal subpopulations and low tissue mRNA levels. Genetic, epigenetic and transcriptional mechanisms that define spatiotemporal expression of the neuropeptide genes have utmost importance for the formation and functions of neural circuits in normal and pathological human brain. 2. 2. Molecular Adaptations in the Endogenous Opioid System in Human and Rodent Brain

    Författare :Muhammad Zubair Hussain; Georgy Bakalkin; Anita Hansson; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Endogenous Opioid System; Dynorphins; Lateralization; Alcohol Dependence; Emotions; Traumatic Brain Injury; Anterior Cingulate Cortex; Dorsal Striatum; Neuroscience; Neurovetenskap;

    Sammanfattning : The aims of the thesis were to examine i) whether the endogenous opioid system (EOS) is lateralized in human brain areas involved in processing of emotions and pain; ii) whether EOS responses to unilateral brain injury depend on side of lesion, and iii) whether in human alcoholics, this system is involved in molecular adaptations in brain areas relevant for cognitive control of addictive behavior and habit formation.The main findings were that (1) opioid peptides but not opioid receptors and classic neurotransmitters are markedly lateralized in the anterior cingulate cortex involved in processing of  positive and negative emotions and affective component of pain. 3. 3. Mechanisms of Prodynorphin Gene Dysregulation in the Brain of Human Alcoholics

    Författare :Malik Mumtaz Taqi; Georgy Bakalkin; Fred Nyberg; Hallberg Mathias; Wolfgang.H Sommer; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; PHARMACY; FARMACI;

    Sammanfattning : The endogenous opioid system (EOS) including dynorphin opioid peptides and κ-opioid receptor (KOR) plays a critical role in alcohol dependence. Aims of the thesis were to evaluate whether the EOS undergoes molecular adaptations in brain areas involved in cognitive control of addiction in human alcohol dependent subjects, and to analyze the impact of genetic and epigenetic factors on these adaptive changes.
