1. Molecular Adaptations in the Endogenous Opioid System in Human and Rodent Brain
Sammanfattning : The aims of the thesis were to examine i) whether the endogenous opioid system (EOS) is lateralized in human brain areas involved in processing of emotions and pain; ii) whether EOS responses to unilateral brain injury depend on side of lesion, and iii) whether in human alcoholics, this system is involved in molecular adaptations in brain areas relevant for cognitive control of addictive behavior and habit formation. The main findings were that (1) opioid peptides but not opioid receptors and classic neurotransmitters are markedly lateralized in the anterior cingulate cortex involved in processing of positive and negative emotions and affective component of pain. 2. Opioid and non-opioid activities of the dynorphins
Sammanfattning : Endogenous opioid peptides a-neoendorphin, dynorphin A (Dyn A), dynorphin B (Dyn B) and big dynorphin (Big Dyn) consisting of Dyn A and Dyn B, collectively known as dynorphins are derived from the precursor protein prodynorphin (PDYN). Dynorphins regulate pain processing and memory acquisition and modulate reward induced by intake of addictive substances. LÄS MER
3. Spectroscopic studies of dynorphin neuropeptides and the amyloid beta-peptide : The consequences of biomembrane interactions
Sammanfattning : Dynorphin A, dynorphin B and big dynorphin are endogenous opioid neuropeptides. They play an important role in a wide variety of physiological functions such as regulation of pain processing and memory acquisition. Such actions are generally mediated through the κ-receptors. 4. Proteasome targeted therapy in arthritis models : effects on pain and inflammation
Sammanfattning : The ubiquitin proteasome system (UPS) regulates numerous cellular functions by selectively degrading cellular proteins. It controls activation of the transcription factor NF-ĸB which is involved in the expression of pro-inflammatory mediators. The UPS regulates neuronal signalling by controlling levels of synaptic proteins during chronic pain. LÄS MER
5. Mechanisms of Prodynorphin Gene Dysregulation in the Brain of Human Alcoholics
Sammanfattning : The endogenous opioid system (EOS) including dynorphin opioid peptides and κ-opioid receptor (KOR) plays a critical role in alcohol dependence. Aims of the thesis were to evaluate whether the EOS undergoes molecular adaptations in brain areas involved in cognitive control of addiction in human alcohol dependent subjects, and to analyze the impact of genetic and epigenetic factors on these adaptive changes.
