  1. 6. Design & Synthesis of Protein Interacting Affinity Ligands and Protease Inhibitors

    Författare :Charlotta Lindquist; Hans Adolfsson; Fredrik Almqvist; Stockholms universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Organic synthesis; Organisk syntes; organisk kemi; Organic Chemistry;

    Sammanfattning : There is a growing need of both protein drugs and synthetic drugs in the fight against many life-threatening diseases. The first part of this thesis deals with the design and synthesis of high affinity binding ligands for the purification of protein drugs. LÄS MER

  2. 7. Ligand binding and enzyme catalysis studied by molecular dynamics simulations

    Författare :Tomas Hansson; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Molecular biology; molecular dynamics simulation; free energy; linear response; drug design; empirical valence bond; HIV protease; protein tyrosine phosphatase; Molekylärbiologi; Molecular biology; Molekylärbiologi; molekylärbiologi; Molecular Biology;

    Sammanfattning : Molecular dynamics simulations and free energy calculations can be applied to biomolecular systems to predict ligand binding affinities. Combined with hybrid quantum-classical potential energy surfaces, such computations can also be used to probe enzymatic catalysis mechanisms. LÄS MER

  3. 8. Alpha1-Antitrypsin: New insights into the biological role in vitro and in vivo

    Författare :Izabela Nita; Enheten för kroniska inflammatoriska och degenerativa sjukdomar; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES;

    Sammanfattning : Alpha1-Antitrypsin (AAT), an acute-phase glycoprotein produced predominantly in the liver by hepatocytes, but also by macrophages, pulmonary alveolar cells and intestinal epithelial cells, is a member of the SERPIN superfamily (SERine Protease INhibitors), and is one of the major inhibitors of neutrophil elastase in human plasma. Although AAT plays a critical role in regulating inflammation by inhibiting neutrophil serine proteases, recent studies suggest that AAT exhibits much broader anti-inflammatory activity. LÄS MER

  4. 9. Prion processing and propagation in neuronal and dendritic cell culture models

    Författare :Katarina M Luhr; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Prion; scrapie; cathepsin; processing; IFN-gamma; DRG; dendritic cell; cytokine;

    Sammanfattning : Prion diseases are rare neurodegenerative diseases, associated with a conversion of a normal cellular protein, PrPC, into its misfolded and partial protease resistant isoform, PrPSc. These diseases affect both animals and humans and are unique since they can not only be genetic and sporadic, but also transmissible. LÄS MER

  5. 10. Studies on early cellular responses during Epstein-Barr virus infection

    Författare :Xinsong Chen; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :;

    Sammanfattning : The human gamma-herpesvirus Epstein-Barr virus (EBV) has been implicated in the pathogenesis of a broad spectrum of lymphoid and epithelial cell malignancies. A characteristic property of the virus is the capacity to establish a non-productive growthpromoting infection in B-lymphocytes. LÄS MER
