    Sammanfattning : The thesis investigates lung function and the effects of rescue surfactant treatment in mechanically ventilated preterm human infants with respiratory distress syndrome (RDS), and the effects of ventilation at birth on the response to surfactant supplementation in preterm lambs. To assess lung function, the author recorded static pressure-volume (P-V) curves during passive deflation from total lung capacity (TLC) to ambient pressure and measured end-expiratory lung volume (FRC) during mechanical ventilation by tracer gas washout.     Sammanfattning : Hyaluronan is an important component of the lung extracellular matrix, with a high capacity for water immobilization, but information on perinatal changes in the lung hyaluronan concentration and their association with changes in the lung water content is limited. In this study, conducted both in rabbit pups and in human infants, we investigated changes in the hyaluronan concentration and distribution in the lung and in the lung water content after preterm or term birth, and changes produced by common antenatal and postnatal pathological conditions and treatments.     Sammanfattning : In systemic sclerosis (SSc), interstitial lung disease (ILD) and engagement of the vascular system lead to increased morbidity and mortality. The aim of this thesis was to elucidate, in a consecutively included cohort of SSc (limited and diffuse) patients (n = 33), the T cell cytokine profile driving the disease in ILD and to explore the role of matrix metalloproteinase 9 (MMP-9) and its inhibitor: tissue inhibitor of metalloproteinase 1 (TIMP-1) in the extracellular matrix (ECM) degrading process leading to fibrous scarring and honey combing.     Sammanfattning : Nitric oxide (NO) is a mediator of vascular smooth muscle tension that metabolises rapidly in blood. NO delivered by inhalation can therefore be used as a selective pulmonary vasodilator to relieve pulmonary hypertension or to improve oxygenation with no systemic effects. LÄS MER
