  1. 1. Acylated cell-penetrating peptides for nucleic acid delivery

    Författare :Tönis Lehto; Ülo Langel; Eric Johnston; Stockholms universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; cell-penetrating peptides; nucleic acid delivery; neurokemi med molekylär neurobiologi; Neurochemistry with Molecular Neurobiology;

    Sammanfattning : In recent decades many new methods have been developed to cure or treat genetical disorders such as cancer, viral infections or inheritable diseases. The problem is that the nucleic acids and their synthetic analogs, oligonucleotides, are not able to cross the cell membrane due to their physicochemical properties like high negative charge and size. 2. 2. Nucleic acid based therapeutic approaches

    Författare :Joacim Elmén; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :;

    Sammanfattning : Nucleic acid therapy comprises several approaches based on nucleic acid as the active therapeutic component to treat human disease. Antisense and short interfering RNA (siRNA) are two gene-silencing techniques in this category. Based on nucleic acid sequence recognition both techniques inhibit gene expression. LÄS MER

  3. 3. Cell-penetrating peptides targeting glioblastomas for nucleic acid delivery in the blood-brain barrier model

    Författare :Artita Srimanee; Ülo Langel; Ines Neundorf; Stockholms universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Blood-brain barrier model; nucleic acid delivery; glioblastomas; cell-penetrating peptides; neurokemi med molekylär neurobiologi; Neurochemistry with Molecular Neurobiology;

    Sammanfattning : Glioblastoma multiforme is the most aggressive form of malignant brain tumor with poor prognosis. The efficacy of brain cancer treatment by chemotherapeutics is limited by the blood-brain barrier (BBB) which allows less than 2% of the small molecules and blocks almost all the macromolecules to transport into the brain. 4. 4. Characterization of nucleic acid delivery with fatty acid modified cell-penetrating peptide nanoparticle formulations

    Författare :Tõnis Lehto; Ülo Langel; Patrick Midoux; Stockholms universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Transfection; gene delivery; oligonucleotide; complexes; non-viral; in vivo; neurokemi med molekylär neurobiologi; Neurochemistry with Molecular Neurobiology;

    Sammanfattning : Recent advances with techniques used for manipulating gene expression have brought us to an era where various gene therapeutic approaches are becoming common therapeutic tools for many previously incurable diseases. The main factor impeding the wider translation of gene therapy is that the active pharmaceutical ingredients used for interfering with gene expression are based on nucleic acids and synthetic oligonucleotides and such molecules do not readily reach their intracellular targets due to their physicochemical properties and therefore they require delivery vectors to cross the cell membrane. 5. 5. Nucleic acid in gene delivery and gene regulation

    Författare :Pedro Moreno; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :;

    Sammanfattning : The concept of gene therapy, initially attributed to a technology that would allow the correction of inherited genetic disease, has evolved over the years. The realization of the immense technical hurdles to achieve genetic correction led to a broadening of the concept now including the transfer of genetic material whose expression will counteract, mitigate or revert a disease phenotype.
