31. Neurorestorative strategies involving neurogenesis, neuronal precursors and stem cells in animal models of Parkinson's disease
Sammanfattning : The general aims of the thesis is to provide evidence for neurogenesis in the adult substantia nigra pars compacta (SNpc), where the dopamine-producing neurons lost in Parkinson's disease (PD) reside; to optimize methods used for detecting newborn nerve cells in adult brain regions with a low rate of neurogenesis; to explore the mechanism of nerve cell death in animal models of parkinsonism and early degenerative and restorative changes in nigral cell populations; and to develop a therapeutic approach that could translate into a future restorative disease-modifying strategy in PD. 32. Deuterium-L-DOPA : a novel means to improve treatment of Parkinson's disease
Sammanfattning : L-DOPA, the precursor of dopamine, is administered to restore dopamine deficiency in Parkinson´s disease (PD) patients. L-DOPA initially provides a sustained symptomatic relief with superior efficacy as compared to other treatments, while long term treatment is complicated by the gradual emergence of troublesome motor complications i.e. LÄS MER
33. Characterization of dopamine D1 receptor agonists in vivo : implications for the treatment of schizophrenia and Parkinson's disease
Sammanfattning : Dopamine is fundamental in human behavior for movement, cognition and reward. A dysfunctional dopamine system is implicated in several neurological and psychiatric disorders such as Parkinson s disease and schizophrenia. Dopamine mediates its effects through five receptors. LÄS MER
34. The non-human primate as a model of human parkinsonism
Sammanfattning : The motor deficits seen in Parkinson’s disease (PD) are the result of a loss of catecholaminergic neurons from the substantia nigra (SN) and the associated loss of striatal dopamine. Clinical signs become evident once striatal dopamine levels decline by at least 70-80% and nigral cell numbers by 40%, and worsen with increased duration of the disease. LÄS MER
35. Generation of dopamine neurons in vivo and from embryonic stem cells in vitro
Sammanfattning : Mesencephalic dopamine (mesDA) neurons degenerate in patients with Parkinson s disease. In order to develop new treatment strategies for rescuing or replacing degenerating mesDA neurons, an improved understanding of the development and survival of these neurons in the normal brain is required.
