36. Biophysical studies of DNA binding – by the large filament-forming protein Rad51 and the small minor-groove binder Hoechst 33258
Sammanfattning : Mechanistic insight into the nature of DNA-binding ligands is crucial for both drug development as well as understanding more complex biological reactions that take place in the cell. In this Thesis, two rather different DNA-binding molecules are considered: 1. LÄS MER
37. Physics of Viral Infectivity: Energetics of Genome Ejection
Sammanfattning : All viruses that infect bacteria, plant, or animal cells involve a genome (RNA or DNA) that is encapsidated by a rigid protein shell. After delivery of the viral genome into the host cell, new capsid proteins, which are encoded by viral DNA or RNA, are expressed and self-assembled into new viral capsids. LÄS MER
38. Cellular Dyes and Bioactive Peptides - Cell Membrane Interactions and Cellular Uptake
Sammanfattning : Knowledge about mechanisms behind interactions of molecules with biomembranes and cellular uptake is very important for understanding biological processes and for drug design. The work described in this thesis has focused on interactions of cellular dyes and bioactive peptides with cells and the cell membrane. LÄS MER
39. Biological and Biophysical Studies on Ruthenium Complexes
Sammanfattning : Small molecules that interact with DNA through intercalation are attractive candidates for treatments against cancer and microbial infections. Octahedral ruthenium(II) polypyridyl complexes, being substitution-inert, readily synthesized and coordinately saturated, has the potential of being developed in to a new family of drugs. LÄS MER
40. Structure determination and thermodynamic stabilization of an engineered protein-protein complex
Sammanfattning : The interaction between two 6 kDa proteins has been investigated. The studied complex of micromolar affinity (Kd) consists of the Z domain derived from staphylococcal protein A and the related protein ZSPA-1, belonging to a group of binding proteins denoted affibody molecules generated via combinatorial engineering of the Z domain.
