1. Molecular mechanisms in cervical carcinogenesis : Studies of clonality, somatic genetic alterations and human papillomavirus variants in cervical pre-invasive and invasive cancer
Sammanfattning : Cervical cancer derives from a series of pre-invasive cervical neoplastic lesions termed CIN I-III (Cervical Intraepithelial Neoplasia) via typical multiple-step processes. In contrast to premalignant disorders in other organs, different degrees of CIN possess a distinct biological behaviour where a proportion spontaneously regress or persist, whereas some progress to invasive cancer. 2. Delection mapping of human 3p in major epithelial types of cancer and fine localization of candidate tumor suppressor genes
Sammanfattning : Allele loss and deletion mapping using microsatellite markers and the detection of homozygous deletions represented until now the most powerful method to localize potential TSGs. Loss of heterozygosity (LOH) involving several chromosome 3p regions accompanied by chromosome 3p deletions are detected in almost 100% of renal cell carcinoma (RCC), small (SCLCs) and more than 90% of non-small (NSCLC) cell lung cancers. 3. A microcell hybrid based elimination test to identify human chromosome 3 regions that antagonize tumor growth
Sammanfattning : Deletions at multiple sites on the short arm of chr3 have been detected in many different types of human tumors. Each of the deleted regions is expected to harbor a tumor antagonizing/suppressor gene(s) (TSG). The earlier studies showed that the tumorigenicity of malignant cells could be suppressed by introducing a single human chr3. LÄS MER
4. Genomic changes defining the progression of human colorectal and cervical tumors
Sammanfattning : Defining changes during the carcinogenesis and progression of tumors is a major way to obtain a better understanding of the mechanisms of cancer development. We therefore investigated the carcinogenesis process in the colon-rectum and in the uterine cervix by different cell chemical, immunohistochemical and cytogenetic methods. LÄS MER
5. Characterization of a putative tumor suppressor region identified by the elimination test on human 3p21.3
Sammanfattning : An experimental system, called the elimination test (Et), was developed to identify tumor growth antagonizing genes. Chromosome 3 containing microcell hybrids (MCHs) were passaged through SCID mice and regular deletion of the short arm of chromosome 3 (3p) were found by cytogenetic and molecular methods. LÄS MER
