1. Defining RCE1 and ICMT as Therapeutic Targets in K-RAS-induced Cancer
Sammanfattning : CAAX proteins, such as the RAS and RHO proteins, are recognized by a specific CAAX motif at the carboxyl terminus, which undergoes posttranslational modifications. First, a lipid group is attached to the cysteine (the “C”) of the CAAX motif by farnesyltransferase (FTase) or geranylgeranyltransferase-I (GGTase-I); second, the –AAX are removed by RAS converting enzyme 1 (RCE1); and third, the cysteine is methylated by isoprenylcysteine carboxyl methyltransferase (ICMT). 2. The Importance of Isoprenylation and Nf1 Deficiency in K-RAS-induced Cancer
Sammanfattning : The RAS and RHO family proteins contribute to tumorigenesis and metastasis and belong to a family of so called CAAX proteins. The membrane targeting and proper function of CAAX proteins are dependent on posttranslational isoprenylation by farnesyltransferase (FTase) or geranylgeranyltransferase type I (GGTase-I). LÄS MER
3. The clinical use of genetic analyses in colorectal cancer
Sammanfattning : Background: Colorectal cancer (CRC) is a common global disease, with a mortality rate of almost 50%. Prognosis is mainly based on the TNM classification. Surgical interventions have the potential of being curative in patients with stage I-III CRC. Adjuvant treatment with chemotherapy enhances the survival rate, especially in stage III cancer. LÄS MER
4. On the clinical value of genetic analysis in colorectal cancer patients
Sammanfattning : In Sweden, the incidence of colorectal cancer (CRC) is about 5000 new cases every year and this number has not changed markedly during the past twenty years. 90-95% of all CRC appear spontaneously, but there are also inherited forms of CRC (5-10%) of which the two most common are familiar adenomatous polyposis (FAP) and hereditary non-polyposis colorectal carcinoma (HNPCC). 5. Computer-aided drug design approaches in developing anti-cancer inhibitors
Sammanfattning : The thesis entitled “computer-aided drug design approaches in developing anti-cancer drug” is divided into a total of six chapters. In the first chapter, an overview of drug discovery and development are introduced. LÄS MER
