  1. 1. The Importance of Isoprenylation and Nf1 Deficiency in K-RAS-induced Cancer

    Författare :Anna-Karin Sjögren; Göteborgs universitet; []
    Nyckelord :FTase; GGTase-I; isoprenylation; K-RAS; NF1; CAAX proteins; lung cancer; AML;

    Sammanfattning : The RAS and RHO family proteins contribute to tumorigenesis and metastasis and belong to a family of so called CAAX proteins. The membrane targeting and proper function of CAAX proteins are dependent on posttranslational isoprenylation by farnesyltransferase (FTase) or geranylgeranyltransferase type I (GGTase-I). LÄS MER

  2. 2. Defining the role of CAAX protein proteolysis and methylation in the pathogenesis and treatment of progeria

    Författare :Xue Chen; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :;

    Sammanfattning : Hutchinson-Gilford progeria syndrome (HGPS) is a rare childhood disease characterized by failure to thrive, bone abnormalities, hair loss, and a shortened life span due to cardiovascular disease, and nuclear shape abnormalities in cultured cells. HGPS is caused by de-novo mutations in LMNA, the gene encoding prelamin A. LÄS MER

  3. 3. Statins, Lipids, and Mutations: Consequences for the Heart and Immune System

    Författare :Emil G. Ivarsson; Göteborgs universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; CAAX Proteins; Statins; RHO Proteins; Dilated Cardiomyopathy;

    Sammanfattning : CAAX proteins are a group of proteins that undergo a three-step protein maturation process that renders the proteins carboxyl-terminus hydrophobic and prone to localize to cellular membranes, where they have their primary function. The first step in this process is called prenylation, which is the covalent attachment of a lipid, either a 15-carbon farnesyl or a 20-carbon geranylgeranyl lipid, to the carboxyl-terminal cysteine residue by the enzymes farnesyltransferase (FTase) and geranylgeranyltransferase type I (GGTase-I), respectively. 4. 4. Defining RCE1 and ICMT as Therapeutic Targets in K-RAS-induced Cancer

    Författare :Annika Wahlström; Göteborgs universitet; []
    Nyckelord :RCE1; ICMT; K-RAS; CAAX proteins; MPD; lung cancer;

    Sammanfattning : CAAX proteins, such as the RAS and RHO proteins, are recognized by a specific CAAX motif at the carboxyl terminus, which undergoes posttranslational modifications. First, a lipid group is attached to the cysteine (the “C”) of the CAAX motif by farnesyltransferase (FTase) or geranylgeranyltransferase-I (GGTase-I); second, the –AAX are removed by RAS converting enzyme 1 (RCE1); and third, the cysteine is methylated by isoprenylcysteine carboxyl methyltransferase (ICMT).
