16. A biophysical study of nucleic acid interactions with analogues and drugs
Sammanfattning : work presented in this thesis concerns studies on the physicochemical nature of interactions between nucleic acids and small ligands. The outcome of such studies can yield insights at a molecular level into the physiological mechanisms of action of biologically active nucleic-acid binding molecules. LÄS MER
17. Molecular regulation and analysis of neural stem and cancer cell characteristics
Sammanfattning : Every single cell within an organism is constantly facing the demanding task of preserving the integrity of its genomic material to secure proper function and clearance from disease. To accomplish this cells rely on DNA repair mechanisms that are well- conserved throughout evolution and highly specialized in removing particular types of DNA damage. LÄS MER
18. Novel FISH methods to unveil genome architecture
Sammanfattning : The genome consists of incredibly long DNA strands that encode all the vital information for the cell to function. The DNA inclusion in the very tight nuclear space and, simultaneously, the establishment of a hierarchical organization of the chromatin that favors transcription of certain genes over others, sparked a long-lasting quest to understand the design principles that govern genome architecture. 19. The Light-up Probe
Sammanfattning : The work presented in this thesis concerns the development of light-up probes and their potential applications in PCR based molecular diagnostics. The light-up probe is a nucleic acid analog (here PNA) oligomer linked to an asymmetric cyanine dye (here thiazole orange) and it combines the excellent hybridization properties of PNA with the large fluorescence enhancement of the dye upon binding to DNA. 20. Gene expression profiling with real-time PCR
Sammanfattning : The sequence of the entire human genome is available today, and this information can be used to understand, characterize, and treat complex diseases such as cancer. Many cellular functions related to survival, growth and differentiation are reflected in the gene expression pattern, and the ability to quantify transcription levels of specific genes is, and always has been, central to research on gene function.
