1. Structural and Biophysical Studies of Nucleic Acids
Sammanfattning : This thesis is based on six research publications concerned with (i) study of the molecular structures and dynamics of modified nucleosides; (ii) investigation of the effect of incorporation of modified nucleosides on the structure of DNA; (iii) examination of the effect of the sugar modifications on the pseudo-aromatic properties (pKa) of the nucleobases; (iv) analysis of the effect of the CH-π interactions on the relative stability of the DNA-RNA hybrid duplexes. The structural stability of the nucleic acids as well as their behavior in molecular recognition is dominated by hydrogen bonding and stacking interactions beside other non-covalent interactions. 2. Studies on Nucleic Acids – Structure and Dynamics
Sammanfattning : This thesis is based on six papers, Papers I-VI, focusing on the interplay between the stabilizing elements of nucleic acids self-assembly; hydrogen bonding, stacking and solvent effects. In Paper I we investigate how the substitution of the O4' for CH2 in the sugar moiety of adenosine (2'-deoxyaristeromycin) at the A6 position of the Dickerson-Drew dodecamer makes the two modified bases exist in a dynamic equilibrium between Hoogsteen and Watson-Crick base pairing in the NMR time scale. 3. Studies on nucleic acids chemistry
Sammanfattning : This thesis is divided into three parts: Part 1: Synthesis and conformational analysis of cyclic RNAs, and comparison with lariat RNAs modelling the lariat formed during processing of Group II and Nuclear pre-mRNA. Synthesis of a hexameric lariat RNA model with a tetranucleotidyl "2' → 5' loop" and a dinucleotidyl "3'-tail" has been accomplished. LÄS MER
4. Cell-penetrating peptides, novel synthetic nucleic acids, and regulation of gene function : Reconnaissance for designing functional conjugates
Sammanfattning : Our genome operates by sending instructions, conveyed by mRNA, for the manufacture of proteins from chromosomal DNA in the nucleus of the cell to the protein synthesizing machinery in the cytoplasm. 5. Recognition and Visualization of Nucleic Acids
Sammanfattning : The work presented in this Thesis focuses on the interaction between nucleic acids and small molecules, as well as binding or incorporation of reporter chromophores into nucleic acids.
