  1. 1. Genome-wide structural and functional protein characterization by ab initio protein structure prediction

    Författare :Lars Malmström; Avdelningen för Biomedicinsk teknik; []
    Nyckelord :TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; Biomedicinska vetenskaper; Biomedical sciences; Saccharymyces cerevisiae; Ab initio protein structure prediction; Protein annotation;

    Sammanfattning : Very little is known about a considerable part of all proteins and it is time consuming and expensive to study each individual protein to determine its function, structure and cellular role. Proteins retain structural, functional and sequential characteristics from ancestral proteins and hence two proteins that share a common ancestor, i.e. LÄS MER

  2. 2. Solvation properties of proteins in membranes

    Författare :Anna CV Johansson; Erik Lindahl; Mounir Tarek; Stockholms universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; membrane protein; lipid bilayer; free energy; solvation; insertion; molecular dynamics simulations; protein mass fraction; Theoretical chemistry; Teoretisk kemi; Biochemistry; biokemi;

    Sammanfattning : Knowledge about the insertion and stabilization of membrane proteins is a key step towards understanding their function and enabling membrane protein design. Transmembrane helices are normally quite hydrophobic to insert efficiently, but there are many exceptions with unfavorable polar or titratable residues. LÄS MER

  3. 3. Characterization of Antigenic Properties and High Throughput Protein Purification

    Författare :Johanna Steen; Sophia Hober; Mats Ohlin; KTH; []
    Nyckelord :TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; high throughput; protein purification; sample preparation; antigen solubility; immunogenicity; epitope mapping; antigen design; Bioengineering; Bioteknik; SRA - Molecular Bioscience; SRA - Molekylär biovetenskap;

    Sammanfattning : To understand the cellular processes, knowledge of the localization and function of proteins are essential. There are several high throughput ventures examining the human proteome. However, there are some bottlenecks in these ventures. For example the production and expression of soluble proteins for analysis. LÄS MER

  4. 4. Microdialysis Sampling, Electrochemical Detection and Mass Spectrometry of Carbohydrates in Monitoring Bioprocesses

    Författare :Nelson Torto; Centrum för analys och syntes; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; monosaccharides; membrane; IPED; ion spray; probe internal geometry; extraction fraction; electrochemical oxidation; electrochemical detection; disaccharides; desalting; carbohydrates; bioprocess monitoring; Cu; APCI; Au; MS; oligosaccharides; PAD; PEI; Analytical chemistry; Analytisk kemi;

    Sammanfattning : This thesis addresses; (1) the microdialysis experiment, application and methodology to maximise its performance when sampling from enzymatic bioprocesses, (2) electrochemical detection and electrochemical oxidation of carbohydrates on Au as well as Cu electrodes in alkaline solutions, and (3) coupling and maximising the performance of a microdialysis sampling based high performance anion exchange chromatography-integrated pulsed electrochemical detection-mass spectrometry system. Studies were carried out to investigate the effect of temperature, analyte as well as biomatrix complexity, analyte concentration, permeability factors, protein(enzyme)-membrane interaction and membrane material on the performance of microdialysis sampling. 5. 5. Improved Techniques for Protein Analysis Focusing on Membrane Proteins and Hydrophobic Peptides

    Författare :Theres Redeby; Åsa Emmer; Jonas ergquist; KTH; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Integral membrane proteins; Hydrophobic peptides; Sample handling; MALDI-TOF-MS; Structured sample support; CE; fraction collection; microchannel; Analytical chemistry; Analytisk kemi;

    Sammanfattning : In this thesis, the vital cell functions performed by integral membrane proteins (IMPs) are briefly discussed. Such proteins are under-represented in most protein studies due to the hydrophobic nature of IMPs, which seriously complicate their solubilization, sample handling, preparation, separation and analysis.
