  1. 21. Expression of multidrug resistance genes and proteins and effect of selenite in anthracycline-resistant human tumor cell lines

    Författare :Kerstin Jönsson Videsäter; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Multidrug resistance; cross-resistance profile; ATP-bioluminescence assay; pglycoprotein; MRP1; selenium; thioredoxin reductase; glutathione reductase.;

    Sammanfattning : Treatment failure due to development of drug resistance is a serious complication in cancer treatment and the major cause of death in these patients. This thesis has focused on to gain knowledge about the mechanisms of multidrug resistance and to establish in vitro methods to characterize the resistant tumor cell phenotype. LÄS MER

  2. 22. Modelling and prediction of drug transport processes with experimental and calculated molecular properties : A multivariate approach

    Författare :Thomas Österberg; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pharmacy; FARMACI; PHARMACY; FARMACI; farmaceutisk farmakologi; Pharmaceutical Pharmacology;

    Sammanfattning : Less than 2% of the lead compounds generated by the pharmaceutical industry enter the exploratory drug-development phase, from which point they stand only a 10% chance of becoming a commercial medicine. A large proportion of the compounds fail due to poor biopharmaceutical properties, such as permeability and solubility. LÄS MER

  3. 23. Intestinal permeability and presystemic metabolism of R/S-verapamil and fluvastatin

    Författare :Rikard Sandström; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pharmacy; FARMACI; PHARMACY; FARMACI; Biopharmaceutics; biofarmaci;

    Sammanfattning : The rate and extent of absorption of drugs from the gastrointestinal tract into the systemiccirculation is defined as the bioavailability. The bioavailability is influenced by a numberof pharmaceutical, pharmacokinetic, biochemical and physiological factors. Two key-variables are the intestinal permeability and the presystemic metabolism. LÄS MER

  4. 24. Pharmacogenetics and Antipsychotic Treatment in Schizophrenia with Special Focus on Adverse Drug Reactions

    Författare :Arzu Gunes; Marja-Liisa Dahl; Maria Gabriella Scordo; Kristina I. Melkersson; Jürgen Brockmöller; Uppsala universitet; []
    Nyckelord :Medicine; Pharmacogenetics; schizophrenia; antipsychotic treatment; adverse effects; clinical outcome; Medicin;

    Sammanfattning : Genetically determined differences in drug metabolism and disposition and drug targets play a pivotal role in the interindividual variability in the clinical outcome of antipsychotic treatment. The aim of this thesis was to study the impact of polymorphisms in genes involved in the pharmacokinetics and pharmacodynamics of antipsychotics, with special focus on their extrapyramidal and metabolic adverse effects. 5. 25. Heterogeneity of leukemic cells with regard to daunorubicin accumulation and efflux : potential role in drug resistance

    Författare :Eva Knaust; Linköpings universitet; []
    Nyckelord :Drug resi stance. acute myeloid leukemia; flow cytometry; gene expression; transport kinetics; apoptosis; MEDICINE; MEDICIN;

    Sammanfattning : Cellular resistance to chemotherapy is a major cause of treatment failure in acute myelocytic leukemia (AML) and other malignancies. Drug resistance is multifactorial; the most studied mechanism being multidrug resistance (MDR), mediated by the P-glycoprotein (PPG), a transmembrane transport protein with a pump function, encoded by the mdr 1 gene.
