6. On the role of cytochrome P450 3A4 in the metabolism of cholesterol and bile acids
Sammanfattning : The origin of 4beta-hydroxycholesterol, a major oxysterol in human circulation, has previously been unknown. In this thesis, the formation and elimination of this compound are investigated by various techniques. The biological effects of 4beta-hydroxycholesterol are also discussed. LÄS MER
7. Mitochondrial disease in children : from clinical presentation to genetic background
Sammanfattning : Mitochondrial disorders are amongst the most common groups of inborn errors of metabolism. They are caused by deficiencies in the final pathway of the cellular energy production, the mitochondrial respiratory chain. LÄS MER
8. Crisis in Energy Metabolism - Mitochondrial Defects and a New Disease Entity
Sammanfattning : Impairment of energy metabolism may be associated with severe implications for affected individuals since all fundamental cell functions are energy-dependent. Disorders of energy metabolism are often genetic and associated with defects in the oxidative phosphorylation in mitochondria. LÄS MER
9. Monitoring of antiepileptic drugs using microdialysis : methodological and clinical aspects
Sammanfattning : In therapeutic monitoring of antiepileptic drugs (AEDs) the level of unbound drug at the sampling site should ideally equal the unbound concentration at receptor sites. Microdialysis enables direct sampling of the unbound drug concentration and avoids some of the difficulties encountered with ex vivo protein binding. LÄS MER
10. 24s-Hydroxycholesterol : studies on regulatory mechanisms behind its formation in the brain and its potential use as a marker for neurodegeneration
Sammanfattning : Cholesterol 24-hydroxylase (CYP46A1) belongs to the cytochrome P450 super family and is responsible for conversion of cholesterol to the oxysterol 24S- hydroxycholesterol (24S-OHC). This structural modification allows 24S-OHC to traverse the blood brain barrier and this pathway is the major one for elimination of cholesterol from the mammalian brain. LÄS MER
