  1. 1. Cytochrome P450 Enzymes in Bile Acid Biosynthesis and Fatty Acid Metabolism : Studies on Members of the Porcine CYP4A and CYP8B Subfamilies

    Författare :Kerstin Lundell; Kjell Wikvall; Ronnie Hansson; Stefan Alexson; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pharmaceutical biochemistry; cytochrome P450; bile acids; fatty acids; CYP4A; CYP8B; genome; porcine; hyocholic acid; Farmaceutisk biokemi; Pharmaceutical biochemistry; Farmaceutisk biokemi; farmaceutisk biokemi; Pharmaceutical Biochemistry;

    Sammanfattning : The present investigation is devoted to studies on porcine members of the cytochrome P450 4A (CYP4A) and CYP8B1 subfamilies, which are involved in bile acid biosynthesis and fatty acid metabolism. Hyocholic acid is considered to fulfil the requirements for trihydroxy bile acids in the domestic pig (Sus scrofa) in the absence of cholic acid. LÄS MER

  2. 2. On the regulation of hepatic cholesterol and bile acid metabolism in gallstone patients

    Författare :Carl-Gustaf Hillebrant; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Bile acid synthesis; biliary lipids; cholesterol synthesis; cholesterol 7a-hydroxylase; cholesterol synthesis inhibitor; chenodeoxycholic acid; cholestyramine; 7a-hydroxy-4-cholesten-3-one; HMG CoA reductase; lathosterol; lipoproteins; lithocholic acid; plasma apheresis; ursodeoxycholic acid;

    Sammanfattning : The liver plays an important role in the regulation of cholesterol homeostasis, therefore also in the pathogenesis of arteriosclerosis and gallstone disease. The quantitatively most important way by which cholesterol is removed from the human body is via the bile, either as free cholesterol or after transformation to bile acids. LÄS MER

  3. 3. Studies on the regulatory role of cholic acid in cholesterol metabolism

    Författare :Charlotte Murphy; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :;

    Sammanfattning : Cholesterol is essential for normal growth and development in both man and mouse. However, excess cholesterol can be harmful leading to diseases such as atherosclerosis and gallstones. Once formed cholesterol cannot be catabolised by the body and must therefore be converted to other compounds for excretion. LÄS MER

  4. 4. Use of primary human hepatocytes for the elucidation of bile acid synthesis

    Författare :Ewa C S Ellis; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :;

    Sammanfattning : In the present investigation we cultured primary human hepatocytes on matrigel under serum-free conditions and studied bile acid formation. The following observations were made: · The major steroid products excreted from primary human hepatocytes into the culture medium were cholic acid, CA (70%) and chenodeoxycholic acid, CDCA (25%) conjugated with glycine or taurine. 5. 5. Studies on cholesterol homeostasis by modulation of lipoproteins and bile acids

    Författare :Lisa-Mari Nilsson; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Cholesterol; lipoproteins; bile acids; cholestyramine; chenodeoxycholic acid; CYP7A1; HNF4alpha; PCSK9; hepatocytes; ethanol; atherosclerosis; ApoE; thyroid hormone receptor;

    Sammanfattning : Excess plasma cholesterol, in particular high levels of LDL cholesterol, is a major risk factor for the development of atherosclerosis. The liver has a central role in the regulation of cholesterol metabolism: i) a major part of the cholesterol present in the body is synthesized in the liver; ii) the level of plasma lipoproteins is regulated by liver uptake and secretion of these particles and iii) the liver is essential for elimination of cholesterol from the body as such or by its conversion into bile acids.
