11. Rational design of novel cell-penetrating peptides
Sammanfattning : Although cell-penetrating peptides (CPPs) have been an active field of research for over 20 years there are still properties such as efficiency and stability that must be improved for CPPs to reach their full potential as delivery vectors. In this thesis two peptides were designed for non-covalent oligonucleotide delivery, in an attempt to overcome two major issues hampering wider use of CPPs, namely low stability and endosomal entrapment. 12. Cell-penetrating peptides targeting glioblastomas for nucleic acid delivery in the blood-brain barrier model
Sammanfattning : Glioblastoma multiforme is the most aggressive form of malignant brain tumor with poor prognosis. The efficacy of brain cancer treatment by chemotherapeutics is limited by the blood-brain barrier (BBB) which allows less than 2% of the small molecules and blocks almost all the macromolecules to transport into the brain. 13. Acylated cell-penetrating peptides for nucleic acid delivery
Sammanfattning : In recent decades many new methods have been developed to cure or treat genetical disorders such as cancer, viral infections or inheritable diseases. The problem is that the nucleic acids and their synthetic analogs, oligonucleotides, are not able to cross the cell membrane due to their physicochemical properties like high negative charge and size. 14. In-silico design of peptide-based transfection systems, in-vitro validation, and up-take pathways investigation
Sammanfattning : Cell-penetrating peptide-based transfection systems (PBTS) are a promising group of drug delivery vectors. Cell-penetrating peptides (CPPs) are short cationic peptides that are able of transporting cell non-permeant cargos into different cell types. LÄS MER
15. Cell-penetrating peptide based nanocomplexes for oligonucleotide delivery
Sammanfattning : Oligonucleotide-based drugs hold great promise for the treatment of many types of diseases, ranging from genetic disorders to viral infections and cancer. The problem is that efficient delivery across the cell membrane is required for oligonucleotides to have their desired effect. Cell-penetrating peptides (CPPs) provide a solution to this problem.
