6. Stimulation of insulin secretion independently from changes in cytosolic free Ca²+-concentration : studies with imidazolines and inositol polyphosphates
Sammanfattning : Insulin secretion from the islets of Langerhans is regulated by changes in glucose concentration. Glucose stimulates secretion by two mechanisms. First, it increases cytosolic free Ca2+ concentration ([Ca2+]i) by inhibition of ATP-dependent K+-channels (KATP) and induction of membrane depolarization. LÄS MER
7. Platelet function in diabetes mellitus : relationships to hyperglycaemia, antidiabetic treatment and microangiopathy
Sammanfattning : Diabetes mellitus is associated with accelerated atherosclerosis and increased morbidity and mortality in micro-and macrovascular complications. The metabolic derangements that accompany diabetes can adversely influence platelets and vascular endothelial function, which may contribute to the pathogenesis of diabetic angiopathy. LÄS MER
8. Effects of high glucose on platelet activation
Sammanfattning : Diabetes mellitus (DM) is associated with increased cardiovascular mortality and morbidity, due to diabetic angiopathy. Hyperglycaemia is one of the factors that may cause platelet dysfunction in diabetic patients. This work investigated the mechanisms underlying hyperglycaemia-induced platelet hyperreactivity and its impact in diabetic patients. LÄS MER
9. Prevention of beta-cell dysfunction via targeting novel GPCRs in pancreatic islets
Sammanfattning : The primary aim of this thesis was to investigate the role of G protein coupled receptors (GPCRs) in insulin secretion and beta-cell survival. The second aim was to determine which pathway is involved in insulin release and beta-cell protection via GPCRs. LÄS MER
10. Protein phosphorylation cascades regulating islet beta-cell insulin exocytosis through glucose sensing and drug signaling : with special focus on serine/threonine protein phosphates and hypoglycemic sulfonylureas
Sammanfattning : In human type 2 diabetes, loss of glucose-sensitive insulin secretion from the pancreatic -cell is an early pathogenetic event. Initially, this may be compensated for by stimulating insulin secretion by sulfonylurea drugs. LÄS MER
