Co-stimulation has proven to be crucial for therapeutic effectiveness of CAR T cells and remarkable clinical response rates have been achieved with second generation CD19-directed CAR T cells containing either a CD28 or 4-1BB co-stimulatory domain for the treatment of B cell malignancies.
