1. Molecular principles of protein stability and protein-protein interactions
Sammanfattning : Proteins with highly specific binding properties constitute the basis for many important applications in biotechnology and medicine. Immunoglobulins have so far been the obvious choice but recent advances in protein engineering have provided several novel constructs that indeed challenge antibodies. LÄS MER
2. Antiadhesive agents targeting uropathogenic Escherichia coli : Multivariate studies of protein-protein and protein-carbohydrate interactions
Sammanfattning : This thesis describes studies directed towards development of novel antiadhesive agents, with particular emphasis on compounds that prevent attachment of bacteria to a host-cell. Three different proteins involved in the assembly or function of adhesive pili in uropathogenic Escherichia coli have been targeted either by rational structure based design or statistical molecular methods. 3. Structure determination and thermodynamic stabilization of an engineered protein-protein complex
Sammanfattning : The interaction between two 6 kDa proteins has been investigated. The studied complex of micromolar affinity (Kd) consists of the Z domain derived from staphylococcal protein A and the related protein ZSPA-1, belonging to a group of binding proteins denoted affibody molecules generated via combinatorial engineering of the Z domain. 4. NMR Studies of Bacterial Albumin-Binding Modules
Sammanfattning : Peptostreptococcus magnus and group C/G streptococci are Gram-positive cocci that are part of the normal flora and reside in more or less the same locations in the host. Protein G of group C/G streptococci was originally described as binding IgGFc of several species and later shown to also bind human serum albumin and albumins from various animal species. LÄS MER
5. Properties of Protein and Polymer Systems
Sammanfattning : Interactions of proteins with other proteins, with polymers and with surfaces are of great practical importance within for instance protein purification, drug delivery, and food technology. The aim of this work is to increase the understanding of protein-protein, protein-polyelectrolyte, and protein-surface interactions, particularly concerning the role of electrostatic interactions in these systems.
