    Sammanfattning : The aims of the studies described in this thesis were to investigate the pharmacokinetics of and cellular resistance to chemotherapy as causes of treatment failure in childhood acute lymphoblastic leukemia (ALL).Leukemic cells from 370 children with newly diagnosed ALL were tested by the Fluorometric Microculture Cytotoxicity Assay to measure their resistance to each of ten standard cytotoxic drugs. 2. 2. Circumventing drug resistance : Studies exploring the expediency of a total cell kill chemoresistance assay

    Författare :Bertil Jonsson; Uppsala universitet; []
    Nyckelord :Medical sciences; drug resistance; drug resistance assay; clinical correlations; cancer; human; MEDICIN OCH VÅRD; MEDICINE; MEDICIN; klinisk farmakologi; Clinical Pharmacology;

    Sammanfattning : Cellular resistance to anti-cancer drugs is considered to be the major cause of treatment failure in clinical practice. The present studies have therefore focused on ways to circumvent resistance as evaluated by an in vitro chemoresistance assay. LÄS MER

  3. 3. Exploring Cancer Drugs In Vitro and In Vivo : With Special Reference to Chemosensitivity Testing and Early Clinical Development

    Författare :Anne von Heideman; Peter Nygren; Bengt Tholander; Rolf Larsson; Seija Grénman; Uppsala universitet; []

    Sammanfattning : The aims of this thesis were to investigate the utility of in vitro drug sensitivity testing to optimize the use of cancer chemotherapy and to assess the properties of a new cancer drug in a phase I clinical trial. Tumour cells from patients were analysed with the short-term Fluorometric Microculture Cytotoxicity Assay (FMCA). LÄS MER

  4. 4. Acute Lymphoblastic Leukaemia in Adult Patients : Studies of Prognostic Factors, Treatment Results and in vitro Cellular Drug Resistance

    Författare :Helene Hallböök; Bengt Smedmyr; Elin Lindhagen; Lorentz Brinch; Uppsala universitet; []

    Sammanfattning : Treatment results and clinical characteristics in adult acute lymphoblastic leukaemia (ALL) were evaluated regarding three issues: a new treatment with cytarabine up-front, stem cell transplantation and a comparison between adult and paediatric treatment protocols. All studies were conducted on a national basis. LÄS MER

  5. 5. Clinical and Experimental Studies in Chronic Myeloid Leukemia : Studies of Treatment Outcome, In Vitro Cellular Drug Resistance and Gene Expression

    Sammanfattning : The aims of the studies described in the thesis were to investigate different treatment strategies in chronic myeloid leukemia (CML) patients. Furthermore, activity of imatinib was investigated by in vitro cytotoxicity assay, and the gene expression pattern in interferon treated patients. LÄS MER
