  1. 1. Blood-Brain Barrier Transport of Drugs Across Species with the Emphasis on Health, Disease and Modelling

    Författare :Karin Tunblad; Margareta Hammarlund-Udenaes; Niclas Jonsson; Elizabeth CM de Lange; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pharmaceutical biosciences; Blood-brain barrier; Disease; Microdialysis; Modelling; Transport; Pharmacokinetics; NONMEM; Farmaceutisk biovetenskap; Biopharmacy; Biofarmaci;

    Sammanfattning : The transport of drugs across the blood-brain barrier (BBB) has been investigated in different species using morphine and morphine-6-glucuronide (M6G) as model compounds. The influence of probenecid on the BBB transport of morphine and M6G was investigated, and the consequences of meningitis and severe brain injury on the concentrations of morphine in the brain were examined. 2. 2. Mechanistic Based Pharmacokinetic-Pharmacodynamics models for Drug Interactions and Disease Population Predictions

    Författare :Luna Prieto Garcia; Erik Sjögren; Aleksandra Galetin; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Physiologically-Based Pharmacokinetics; Pharmacokinetics; Pharmacodynamics; Disease Modelling; Drug Interactions; Statins; Mechanistic Modelling; Farmakokinetik och läkemedelsterapi; Pharmacokinetics and Drug Therapy;

    Sammanfattning : Therapeutic dose of a medication refers to the quantity of a drug required to produce a pharmacological effect without causing unacceptable adverse events. Dose selection in the clinical setting is not straight forward due to various factors, including specific patient factors such as age, sex, weight, genetic variants and renal/hepatic function, as well as external factors such as food and co-medication, all of which can influence the efficacy and safety of a drug. 3. 3. Semi-mechanistic models of glucose homeostasis and disease progression in type 2 diabetes

    Författare :Steve Choy; Maria Kjellsson; Mats Karlsson; Willem de Winter; Andrea Mari; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; pharmacokinetics; pharmacodynamics; pharmacometrics; glucose homeostasis; insulin; type 2 diabetes; obesity; weight; visceral adipose tissue; HbA1c; non-linear mixed effects; modelling; disease progression; ZDSD rats; Farmaceutisk vetenskap; Pharmaceutical Science;

    Sammanfattning : Type 2 diabetes mellitus (T2DM) is a metabolic disorder characterized by consistently high blood glucose, resulting from a combination of insulin resistance and reduced capacity of β-cells to secret insulin. While the exact causes of T2DM is yet unknown, obesity is known to be a major risk factor as well as co-morbidity for T2DM. LÄS MER

  4. 4. Characterisation of inputs and outputs of striatal medium spiny neurons in health and disease

    Författare :Marko Filipović; Jeanette Hellgren Kotaleski; Arvind Kumar; Gilad Silberberg; Ad Aertsen; Andrew Sharott; KTH; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; basal ganglia; striatum; medium spiny neurons; subthalamic nucleus; globus pallidus; Parkinson s disease; in vivo; oscillations; spectral analysis; response variability; networks; simulations; modelling; Numerical Analysis; Numerisk analys; Computer Science; Datalogi; Biological and Biomedical Physics; Biologisk och biomedicinsk fysik;

    Sammanfattning : Striatal medium spiny neurons (MSNs) play a crucial role in various motor and cognitive functions. They are separated into those belonging to the direct pathway (dMSNs) and the indirect pathway (iMSNs) of the basal ganglia, depending on whether they express D1 or D2 type dopamine receptors, respectively. Sammanfattning : A challenge for the clinical management of Parkinson's disease (PD) is the large within- and between-patient variability in symptom profiles as well as the emergence of motor complications which represent a significant source of disability in patients. This thesis deals with the development and evaluation of methods and systems for supporting the management of PD by using repeated measures, consisting of subjective assessments of symptoms and objective assessments of motor function through fine motor tests (spirography and tapping), collected by means of a telemetry touch screen device.
