    Författare :Lingquan Deng; Olof Ramström; Javier Rojo; KTH; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Carbohydrates; Glycobiology; Carbohydrate-protein interactions; Perfluorophenylazide PFPA ; Photochemistry; Carbohydrate surface immobilization; Carbohydrate microarrays; Surface plasmon resonance SPR ; Quartz crystal microbalance QCM ; Atomic force microscopy AFM ; Glyconanomaterials; Concanavalin A Con A ; Cyanovirin-N CV-N ; 1-Glycosyl thiols; Mutarotation; 1-S-glycosylation; Stereocontrol;

    Sammanfattning : This thesis mainly deals with the development of photochemical approaches to immobilize carbohydrates on surfaces for glycobiological studies. These approaches have been incorporated into a number of state-of-the-art nanobio-platforms, including carbohydrate microarrays, surface plasmon resonance (SPR), quartz crystal microbalance (QCM), atomic force microscopy (AFM), and glyconanomaterials. 2. 2. Functional studies and engineering of family 1 carbohydrate-binding modules

    Författare :Janne Lehtiö; KTH; []
    Nyckelord :Cellulose-binding; carbohydrate-binding modules; phage display; bacterial surface display; combinatorial protein library; protein engineering; Trichoderma reesei; Staphylococcus carnosus;

    Sammanfattning : The family 1 cellulose-binding modules (CBM1) form a groupof small, stable carbohydrate-binding proteins. These modulesare essential for fungal cellulosedegradation. This thesisdescribes both functional studies of the CBM1s as well asprotein engineering of the modules for several objectives. LÄS MER

    Sammanfattning : Owing to the current efforts to find sustainable alternatives to petrochemical based industries and technologies, enzymatic degradation and valorization of plant biomass has been attracting interest. Due to the complexity of plant biomass, an array of enzymes is required to hydrolyze it, including esterases. LÄS MER

  5. 5. Improving the biocatalyst. Engineering of fungal oxidoreductases and thin-film electrodes for improvement of membraneless biofuel cells and carbohydrate biosensors

    Författare :Roberto Ortiz; Biokemi och Strukturbiologi; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; Cellobiose dehydrogenase; pyranose dehydrogenase; biosensor; enzymatic biofuel cell; carbon nanotubes; gold nanoparticles; direct electron transfer; flow-injection analysis; diazonium salts; self-assembled monolayers;

    Sammanfattning : This thesis presents investigations for the improvement of electrical contact and catalytical properties of two fungal redox enzymes, cellobiose dehydrogenase (CDH) and pyranose dehydrogenase (PDH), with electrodes and the use of the prepared electrodes as carbohydrate biosensors and in enzymatic biofuel cells. The performance of enzyme modified electrodes can be improved in two ways, by modifying the bioelement or by modifying the electrode. LÄS MER
