6. Dealing with damaged lysosomes : Impact of lysosomal membrane stability in health and disease
Sammanfattning : The lysosome is the main unit for degradation and plays important roles in various cellular processes, such as nutrient sensing, cholesterol regulation and cell death. Consequently, altered lysosomal function contributes to, or even causes, several diseases. LÄS MER
7. Metabolic disorders : genes and mechanisms
Sammanfattning : Mitochondrial disorders constitute a subgroup of the large family of rare monogenic diseases called Inborn Errors of Metabolism (IEM), but defects in mitochondrial activity have also been reported in more common and complex pathologies. In the present thesis, I validate new disease-causing variants and study the underlying molecular defects in three different cases of IEM. LÄS MER
8. Non-canonical ATG8 conjugation in ESCRT-driven membrane remodeling processes
Sammanfattning : ATG8 family proteins have the unique ability to conjugate to membrane lipids. Initially identified as a hallmark of autophagy, ATG8 lipidation is emerging as an important regulator of a growing list of non-degradative cellular functions. LÄS MER
9. Short and long-term regulation of autophagy
Sammanfattning : Autophagy is a conserved catabolic pathway triggered by stress conditions in which portions of the cytoplasm, damage organelles, misfolded proteins and intracellular bacteria are delivered and degraded in the lysosome/vacuoles. Thus, an efficient induction and completion of the process is required to ensure a proper homeostasis of the cell. LÄS MER
10. CADASIL : a pure model for studying cerebral small vessel disease
Sammanfattning : Cerebral autosomal dominant arteriopathy with subcortical infarct and leukoencephalopathy (CADASIL) is caused by a mutation on the NOTCH3 gene. The pathological driver behind this disease is the loss of vascular smooth muscle cells (VSMCs) in small blood vessels and subsequent fibrotic thickening of the vessel, causing stenosis. LÄS MER
