  1. 1. Role of Bone Morphogenetic Proteins for Catecholaminergic Neurons in Vivo : Use of the Tyrosine Hydroxylase Locus for Cell-Specific inactivation of Signal Transduction

    Författare :Dmitry Usoskin; Ted Ebendal; Ernest Arenas; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Neurosciences; Tyrosine Hydroxylase; Bone Morphogenetic Proteins BMP ; Transforming Growth Factor-β TGF-β ; tissue-specific knockout; Parkinson s disease; Neurovetenskap; Neurology; Neurologi;

    Sammanfattning : Members of the Transforming Growth factor-β (TGF-β) superfamily and its subclass Bone Morphogenetic Proteins (BMP) play important roles for nervous system development. In order to study the BMP role for catecholaminergic neurons in vivo, we generated three knock-in mice, expressing the transgenes specifically in the targeting cells. LÄS MER

  2. 2. Smad signaling in hematopoietic stem cell biology

    Författare :Göran Karlsson; Avdelningen för molekylärmedicin och genterapi; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Smad7; Smad4; microarray; retroviral-mediated gene expression; conditional knockout; transforming growth factor beta; smad signaling; hematopoiesis; hematopoietic stem cells; Alk5; Medicine human and vertebrates ; Medicin människa och djur ;

    Sammanfattning : Hematopoietic stem cells (HSCs) are primitive tissue-specific somatic stem cells, responsible for the maintenance and replenishment of the bone marrow (BM) and subsequently the entire blood system. The never-ending ability to differentiate into all the mature hematopoietic lineages makes these HSCs attractive candidates for use in future regenerative medicine. LÄS MER

  3. 3. Life without mitochondrial DNA : studies of transgenic mice

    Författare :Jianming Wang; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :mitochondrial disease; mitochondrial DNA mtDNA ; mitochondrial transcripton factor A Tfam ; cre-loxP; conditional knockout; transgenic; mitochondrial cardiomyopathy; apoptosis;

    Sammanfattning : Mitochondrial DNA (mtDNA) is a closed circular DNA genome that resides in the mitochondrial network. Mutations of mtDNA cause spontaneous and hereditary disorders known as mitochondrial diseases. Mitochondrial transcription factor A (Tfam) is a key factor for transcription of mtDNA in vitro. 4. 4. The pathophysiology of respiratory chain dysfunction

    Författare :José Pablo Silva; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Mitochondrial disease; mitochondrial DNA mtDNA ; mitochondrial transcription factor A Tfam ; Cre-loxP; conditional knockout; reactive oxygen species ROS ; superoxide; uncoupling protein UCP ; mitochondrial bioenergeties; energy expenditure;

    Sammanfattning : Mutations of mitochondrial DNA cause a variety of clinical syndromes. It is unclear whether impaired oxidative phosphorylation on its own is the main cause of pathology or whether other factors such as secondary metabolic alterations, enhanced formation of reactive oxygen species (ROS) and induction of apoptosis also may contribute to the clinical phenotype. 5. 5. AMP-activated protein kinase : the connection between exercise and type II diabetes

    Författare :Brian R Barnes; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Skeletal muscle; glucose transport; AMP-activated protein kinase; AMPK gamma 3 exercise; Type 2 diabetes; insulin resistance; metabolism;

    Sammanfattning : Skeletal muscle insulin resistance is a hallmark feature of Type 2 diabetes. Physical exercise/muscle contraction elicits an insulin-independent increase in glucose transport and perturbation of this pathway can bypass defective insulin signaling. LÄS MER
