  1. 11. Combinatorial protein engineering applied to enzyme catalysis and molecular recognition

    Författare :Malin Eklund; KTH; []
    Nyckelord :phage display; selection; enzyme; affinity domains; crystal structure; affibody; lipase; protein engineering; protein A; assembly;

    Sammanfattning : The recent development of methods for constructing andhandling large collections (libraries) of proteins, from whichvariants with desired traits can be isolated, hasrevolutionized the field of protein engineering. Key elementsof such methods are the various ways in which the genotypes(the genes) and the phenotypes (the encoded proteins) arephysically linked during the process. LÄS MER

  2. 12. Molecular targets for glucocorticoids in macrophages: cytosolic phospholipase A2 and cytokine formation

    Författare :Karin Gewert; Infektionsmedicin; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; pharmacognosy; Pharmacological sciences; signal transduction; dexamethasone; cytosolic phospholipase A2; glucocorticoids; macrophages; protein kinase C; mitogen-activated protein kinases; TNF-alpha; IL-1beta; protein phosphatases; okadaic acid; pharmacy; toxicology; Farmakologi; farmakognosi; farmaci; toxikologi;

    Sammanfattning : The aim of this investigation has been to study mechanisms by which glucocorticoids inhibit inflammatory processes in vitro. Type IV cytosolic 85 kDa phospholipase A2 (cPLA2) is an important modulator of inflammatory responses that is present in macrophages as well as in other cell types. LÄS MER

  3. 13. Discovery of Novel Macrocyclic Ligands for Difficult Targets : Applications to Natural Product Derived Keap1 Inhibitors

    Författare :Fabio Begnini; Jan Kihlberg; Anders Bach; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Macrocycles; Keap1-Nrf2; Protein-protein interactions; Natural products;

    Sammanfattning : The development of small-molecule ligands for biological targets that possess large, featureless or highly polar binding sites is a challenging task. This thesis is focused on a novel lead generation strategy to identify macrocyclic ligands for difficult-to-drug targets, as well as on the relationship between cell permeability and the conformations of macrocycles. 4. 14. Viperin vs. tick-borne encephalitis virus : mechanism of a potent antiviral protein

    Författare :Kirstin Vonderstein; Anna Överby Wernstedt; Magnus Evander; Sonja Best; Umeå universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Tick-borne encephalitis virus; TBEV; type-I interferon; IFN; Interferon-stimulated gene; ISG; viperin; GBF1; CIAO1; antiviral protein; genome replication; particle release; capsid;

    Sammanfattning : Tick-borne encephalitis virus (TBEV) is a very important virus medically, causing mild or severe encephalitis often with long-lasting sequelae. Treatment of tick-borne encephalitis is limited to supportive care, and antiviral drugs are much needed.The type-I interferon (IFN) system is the first line of host defense against many viruses. 5. 15. Metabolic Engineering of Recombinant Protein Production by Saccharomyces cerevisiae

    Författare :Zihe Liu; Chalmers tekniska högskola; []
    Nyckelord :TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; recombinant protein production; genome sequencing; DNA microarray; insulin precursor; growth rate; UV mutation; yeast; anaerobic electron acceptor; secretory pathway; unfolded protein response; metabolic engineering; systems biology; α-amylase;

    Sammanfattning : The yeast Saccharomyces cerevisiae is a widely used cell factory for the production of fuels, chemicals, and it also provides a platform for the production of many heterologous proteins of medical or industrial interest. In this thesis, random and rational approaches, such as vector design, host engineering, fermentation analysis, UV Mutation, coupled with high-throughput systems biology techniques (including whole genomic sequencing, microarray analysis and flux analysis) and integrated analysis (Reporter feature technique), were employed to engineer cellular properties more effectively and purposefully to construct cell factories for protein production.
