  1. 1. Structural studies on the integral membrane protein, ubiquinol oxidase from Escherichia coli

    Författare :Jeff Abramson; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Biochemistry; crystal structure; ubiquinol oxidase; membrane protein; oxidases; Biokemi; Biochemistry; Biokemi; biokemi; Biochemistry;

    Sammanfattning : Heme-copper oxidases are redox-driven proton pumps that couple the reduction of molecular oxygen to water with the vectorial translocation of protons across the membrane. The proton gradient generated by heme-copper oxidases and the other members of the aerobic respiratory chain is ultimately used to drive the synthesis of ATP. LÄS MER

  2. 2. Experimental and computational exploration of enzyme sequence space

    Författare :Elzbieta Rembeza; Chalmers tekniska högskola; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; high-throughput-screening; protein annotation; enzyme discovery; enzyme sequence space; oxidases;

    Sammanfattning : Millions of enzymes with desirable features or new exciting activities can be found in organisms occupying diverse niches all around the earth. However, enzyme studies tend to be biased towards characterisation of representatives from eukaryotes, model organisms, or disease-causing bacteria. LÄS MER

  3. 3. Amine oxidases in dental pulp : a study with special regard to a semicarbazide-sensitive amine oxidase with catalytic potency towards serotonin

    Författare :Astrid Norqvist-Sjöberg; Lars Oreland; Umeå universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES;

    Sammanfattning : The amine oxidizing capacity of piglet dental pulp tissue has been investigated. At least six separate enzyme activities could be distinguished:- a soluble semicarbazide-sensitive (SSAO) activity towards benzylamine, most likely to be derived from the content of blood plasma within the pulp tissue;- a soluble pulp tissue SSAO activity with most properties in common with the plasma enzyme, however with great efficiency not only regarding benzylamine but also tryptamine;- a tissue bound SSAO activity with activity towards benzylamine, tryptamine, tyramine and ß-phenylethylamine;- a tissue bound semicarbazide-sensitive activity towards serotonin (SSA0-5HT) (a combination of properties never reported before);- a mitochondrially bound and clorgyline-sensitive monoamine oxidase-A (MAO-A) activity with activity towards serotonin;- a mitochondrially bound and deprenyl-sensitive MAO-B activity with activity towards e. 4. 4. Proton transfer in nitric oxide reducing heme-copper oxidases

    Författare :Joachim Reimann; Pia Ädelroth; Peter Brzezinski; Michael Wilson; Stockholms universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; heme-copper oxidases; nitric oxide; proton transfer; electron transfer; proton-transfer pathway; Biochemistry; biokemi;

    Sammanfattning : Heme-copper oxidases (HCuOs) are best known as terminal oxidases in the aerobic respiratory chain, in which they catalyze the reduction of oxygen to water. By receiving protons and electrons from opposite sides of the membrane as well as pumping protons, HCuOs contribute to the electrochemical proton gradient over the membrane that can be used for ATP synthesis. 5. 5. Experimental studies of proton translocation reactions in biological systems : Electrogenic events in heme-copper oxidases

    Författare :Håkan Lepp; Peter Brzezinski; Thomas Nilsson; Stockholms universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; heme-copper oxidase; cytochrome c oxidase; nitric oxide reductase; cbb3-type oxidase; proton pumping; uncoupling; charge-transfer; electrogenic event; flow-flash; Biophysics; Biofysik; Biophysics; biofysik;

    Sammanfattning : Terminal heme-copper oxidases (HCuOs) are transmembrane proteins that catalyze the final step in the respiratory chain - the reduction of O2 to H2O, coupled to energy conservation by generation of an electrochemical proton gradient. The most extensively investigated of the HCuOs are the aa3-type oxidases, to which cytochrome c oxidase (CytcO) belongs, which uses energy released in the O2-reduction for proton pumping. LÄS MER
