    Sammanfattning : Obesity is a major health epidemic worldwide and occurs when long-term excess of energy intake beyond energy utilization leads to an increase in adipose tissue. Energy balance is coordinated by the central nervous system regulation of appetite and adaptive thermogenesis via a complex array of neuropeptides which communicate to the periphery by various circulating measures dictating the utilization of various energy sources such as, carbohydrates, fat, or protein. This thesis focuses on the development of pharmacometric models to characterize and quantify the relationships between drug exposure, circulating and imaging biomarkers, adverse effects, overall survival (OS), and patient-reported outcomes (PROs).   3. 3. Genetic association and risk prediction of breast cancer from an epidemiological and biostatistical perspective

    Sammanfattning : The main purposes of this thesis were to analyse common genetic variation in candidate genes and candidate pathways in relation to breast cancer risk, prognosticators and survival, to develop statistical methods for genetic association analysis for evaluating the joint importance of genes, and to investigate the potential impact of adding genetic information to clinical risk factors for projecting individualised risk of developing breast cancer over specific time periods. In Paper I we studied genetic variation in the estrogen receptor α and epidermal growth factor genes in relation to breast cancer risk and survival. LÄS MER
