  1. 1. Development of Gene Therapy for Hematopoietic Stem Cells using Lentiviral Vectors

    Författare :Niels-Bjarne Woods; Andreas Ooka; Stefan Karlsson; Avdelningen för molekylärmedicin och genterapi; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; embryonic stem cell; Hematopoietic stem cell; lentiviral vector; transduction efficiency; transgene expression; multiple vector copy integration; insertional mutagenesis; NOD SCID mouse; cytogenetics; Genetics; extracellulära vätskor; extracellular fluids; cytogenetik; Genetik; Haematology; Hematologi; Lentivirus : genetics; Human; Genetic Vectors; Hematopoietic Stem Cells : metabolism; Animal; Gene Therapy; Leukemia : therapy; Leukemia : metabolism;

    Sammanfattning : Hematopoietic stem cells are an ideal target for genetic manipulation for the purpose of curing hematological disorders as they have the ability to develop into all blood lineages and to self-renew. In this study we demonstrate that lentiviral vectors, based on HIV-1, can efficiently transfer genes into human hematopoietic progenitor and stem cells as assessed following stem cell transplantation in immune compromised mice. 2. 2. Novel immunotherapies for metastatic melanoma - from mouse models towards clinical trials

    Författare :Elin Forsberg; Göteborgs universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Metastatic melanoma; uveal melanoma; patient-derived xenograft; immune-humanized mouse model; immunotherapy; adoptive T cell therapy; tumor-infiltrating lymphocytes; chimeric antigen receptor T CAR-T cells;

    Sammanfattning : Immunotherapies including checkpoint blockade and adoptive T cell transfer (ACT) show great promise for the treatment of melanoma, with long-term effects in some patients. However, around half of the patients with metastatic malignant melanoma will not be cured with available therapies today, and these patients require other treatment strategies. LÄS MER

  3. 3. Humanized mice as a model to study human immunity

    Författare :Frank Heuts; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :;

    Sammanfattning : Mice with human immune systems have evolved over the last three decades. Over the years, these humanized mouse models have provided us with valuable information on human immunity. Recent developments in recipient mouse strains and engraftment protocols have resulted in models with high level of de novo formation of human immune cells. LÄS MER
