1. Fluorescence Probing of DNA Structures in Biology and Nanotechnology
Sammanfattning : Much has been learnt about DNA since its structure was discovered about half a century ago. Scientists everlasting urge to look beyond and deeper into the intricate machinery of life, has continuously pushed technology forward. LÄS MER
2. DNA precursor biosynthesis-allosteric regulation and medical applications
Sammanfattning : Ribonucleotide reductase (RNR) is a key enzyme for de novo dNTP biosynthesis. We have studied nucleotide-dependent oligomerization of the allosterically regulated mammalian RNR using a mass spectrometry–related technique called Gas-phase Electrophoretic Mobility Macromolecule Analysis (GEMMA). 4. Intelligent Nucleic Acid Interactions with Peptide Nucleic Acid (PNA) and Recombination Enzyme RecA
Sammanfattning : The work presented in this thesis focuses on interactions between nucleic-acids in biological systems, particularly interactions of the promising drug-candidate peptide nucleic acid (PNA), and on protein-DNA intermediates during recombination. Several experimental techniques, mainly circular dichroism and fluorescence spectroscopy, have been used to characterize the interactions of interest. LÄS MER
5. Model Systems for Metal - DNA Interactions. With a Focus on Anticancer Active Platinum(II) Complexes
Sammanfattning : Reactions of trans-dicyanodichloroaurate(III) trans-dibromodicyanoaurate(III), trans-diamminedichlorogold(III), cis-diammine(aqua)chloroplatinum(II), cis-ammineaqua-(cyclohexylamine)chloroplatinum(II), chloro(terpyridine)platinum(II) and chloro-(diethylenetriaminepalldium(II) with dimethyl sulfide, thionucleosides, thionucleotides and thione- and guanine-containing 17-mer oligonucleotides have been investigated by use of conventional and high-pressure stopped-flow spectrophotometry and HPLC technique as a function of ionic strength, pH, temperature and pressure in aqueous solutions. The mechanism and rate of oxidation of dimethyl sulfide by gold(III) complexes depends on the concentration of free halide. LÄS MER
